IL-17在脂多糖致大鼠ALI中的表达及地塞米松的影响

【摘要】:目的:观察IL-17在脂多糖致大鼠ALI模型的表达,并观察早期应用小剂量地塞米松(1mg/kg)对其影响。 方法:将成年健康雄性SD大鼠48只随机分为正常对照组(C组),LPS致ALI组(ALI组),地塞米松组(D组)。腹腔注射脂多糖5mg/kg制作大鼠ALI模型,后立即腹腔注射地塞米松lmg/kg制作D组,C组腹腔注射等剂量的生理盐水。于2h、6h两点各组处死8只大鼠,提取左肺BALF和右肺组织。测定各组肺脏湿/干质量比值(W/D),以及各组肺组织病理形态学变化;ELISA法测定BALF中IL-17、TNF-α水平 结果:1BALF中IL-17含量与C组比较,ALI组2h点IL-17含量无明显差异,6h点IL-17含量显著升高,差异有统计学意义(P0.05);2h点D组IL-17含量与C组及ALI组无差异,6h点IL-17含量较ALJ组减少,但仍高于C组,其与其他两组比较均有统计学意义(P0.05)2BALF中TNF-α含量与C组比较,ALI组TNF-α含量明显增加,并随时间延长而增加,两组差异有统计学意义(P0.05),D组TNF-α含量较ALI组减少,但仍高于C组,其与其他两组比较均有统计学意义(P0.05)。3肺组织W/D C组两时间点差异无统计学意义,ALI组明显升高,各时点与C组相比差异有统计学意义(P0.05),D组较ALI组明显降低,但仍高于C组。4肺组织病理学变化:ALI组两肺出现程度不一的肿胀充血,重者可见散在出血点;光学显微镜可见弥漫性肺泡间隔增厚,部分肺泡萎陷,肺间质弥漫性出血,肺泡腔可见大量炎性细胞浸润及渗液。D组肺组织肿胀充血、肺泡腔炎性细胞浸润及渗液、肺泡间隔增厚及肺泡萎陷程度均较ALI组明显减轻。 结论:IL-17参与了LPS致大鼠ALI过程,6h时显著表达。早期应用小剂量地塞米松能降低大鼠ALI模型BALF中IL-17的表达,可能是地塞米松减轻ALI的另一机制。
[Abstract]:Aim: to observe the expression of IL-17 in rat ALI model induced by lipopolysaccharide, and to observe the effect of low dose dexamethasone (1mg/kg) on it. Methods: Forty-eight adult male SD rats were randomly divided into normal control group (C group), LPS induced ALI group (ALI group) and dexamethasone group (D group). Rat ALI model was established by intraperitoneal injection of lipopolysaccharide (5mg/kg), then dexamethasone lmg/kg was injected intraperitoneally to make group D and group C was intraperitoneally injected with saline of the same dose. At 2 h and 6 h, 8 rats were killed in each group. BALF of left lung and right lung tissue were extracted. The wet / dry weight ratio of lung (W / D) and the pathological changes of lung tissue were measured in each group. The results of IL-17,TNF- 伪 determination in BALF by ELISA: compared with C group, the content of IL-17 in 1BALF group was not significantly different from that in group C at 2h point, but IL-17 content in ALI group was significantly increased at 6h point (P0.05). The content of IL-17 in group D was not different from that in group C and group ALI at 2h, but the content of IL-17 in group D was lower than that in group ALJ at 6 h, but still higher than that in group C. there was significant difference between group D and other two groups (P0.05) the content of TNF- 伪 in 2BALF was higher than that in group C (P0.05). The content of TNF- 伪 in ALI group was significantly higher than that in ALI group, and increased with time. The difference between the two groups was statistically significant (P0.05). The content of TNF- 伪 in), D group was lower than that in ALI group, but still higher than that in C group. Compared with the other two groups, there was statistical significance (P0.05). 3 there was no significant difference in lung tissue at two time points in WR / D C group, ALI group was significantly higher than that in C group (P0.05). Group D was significantly lower than group ALI, but still higher than group C. 4 the pathological changes of lung in group D were higher than those in group C. the swelling and congestion of the two lungs in ALI group were different in degree, and the heavy ones could be seen at the bleeding point. Optical microscope showed diffuse alveolar septum thickening, partial alveolar collapse, diffuse hemorrhage of pulmonary interstitial, infiltration and exudation of a large number of inflammatory cells in alveolar cavity, swelling and congestion of lung tissue, infiltration and exudation of inflammatory cells in alveolar cavity in group D. The thickening of alveolar septum and the degree of alveolar collapse were significantly reduced compared with ALI group. Conclusion: IL-17 was involved in the process of ALI induced by LPS in rats, and was significantly expressed at 6 h. Early application of low dose dexamethasone can reduce the expression of IL-17 in BALF of rat ALI model, which may be another mechanism of reducing ALI by dexamethasone.

Conclusion: IL-17 was involved in the process of ALI induced by LPS in rats, and was significantly expressed at 6 h. Early application of low dose dexamethasone can reduce the expression of IL-17 in BALF of rat ALI model, which may be another mechanism of reducing ALI by dexamethasone.
