神经酰胺对胎鼠肺泡Ⅱ型上皮细胞增殖的影响

【摘要】：取1只妊娠17.5 d的ICR小鼠,将其体内的胎鼠肺泡Ⅱ型上皮细胞(AECⅡ)经分离、纯化、培养、鉴定后,分别加入不同浓度的神经酰胺,培养12、24和48 h后进行MTT试验。在Transwell上构建AECⅡ单层,以10μmol/L的神经酰胺与AECⅡ培养12和24 h,并设置相应对照组,应用伏欧计测定跨上皮电阻值。结果显示,随着神经酰胺染毒浓度的升高和染毒时间的延长,AECⅡ细胞增殖抑制率呈上升趋势。10μmol/L的神经酰胺分别刺激融合的AECⅡ单层12和24 h后,与未处理对照组比较,神经酰胺刺激组的跨上皮电阻值显著下降。
