广西红水河流域人群OBFC1 rs4387287基因多态性与血脂水平的相关性研究

【摘要】:背景:寡核苷酸结合摺叠区1基因(oligonucleotide/oligosaccharide binding fold containing 1 OBFC1)与酵母的Stn1基因具有同源性,并且是直接参与端粒生物功能调节的重要基因之一[1,2]。迄今为止,国内外对OBFC1基因多态性与血脂的相关性研究甚少,为了了解该基因与血脂的关系,我们进行了相关研究。目的:探讨OBFC1 rs4387287多态性与血脂水平的关系,了解该基因对广西红水河流域长寿人群的影响。方法:将2262名研究对象分为四组:长寿组为广西红水河流域的长寿老人,年龄90岁,平均年龄93.27±3.01,共496人;长寿子女组为广西红水河流域的长寿老人的子女,年龄40-75岁,平均年龄59.84±7.81,共723人;当地对照组为广西红水河流域的非长寿家系老人,年龄40-75岁,平均年龄63.42±7.77,共611人;平果对照组为平果地区的非长寿家系老人,年龄40-75岁,平均年龄60.58±7.99,共432人;所选人群均为壮族(参考身份证件)。本次研究主要对身高、体重、血压、血脂、血糖等指标进行测量,应用改良多重酶连接反应(improved multiplex ligation detection reaction,i MLDR)技术对OBFC1 rs4387287位点进行分型,从而了解该基因多态性对血脂水平的影响。结果:1.各临床资料(血脂、血糖和血压)的组间比较长寿组的TG、HDL-C水平低于长寿子女组和当地对照组(均P0.05),但SBP、DBP却出现相反趋势。平果对照组的tc、ldl-c、sbp、血糖水平低于长寿子女组和当地对照组(均p0.05)。2.obfc1rs4387287基因型及等位基因频率分布情况在总人群中,基因型以cc基因型较为多见,频率为71.3%;ca基因型的频率为24.5%;aa基因型的频率为4.2%;c等位基因的频率为83.6%;a等位基因频率为16.4%。a等位基因在长寿组的频率为16.2%;在长寿子女组的频率为15.8%;在当地对照组的频率为15.3%;在平果对照组的频率为19.2%,其中a等位基因在平果对照组的频率高于其余三组(均p0.05)。在当地对照组中,男性cc基因型频率高于同组女性,a等位基因频率低于同组女性(均p0.05)。3.obfc1rs4387287基因型及等位基因对人群血脂水平的影响在平果对照组中,cc基因型的tc水平高于非cc基因型(p=0.014)。性别分层后,在总体男性人群中,cc基因型的hdl-c水平低于非cc基因型(p=0.016);在平果对照组男性中,cc基因型的tc、tg水平高于非cc基因型(p分别为0.008、0.009);在长寿子女组及当地对照组的男性群体中,cc基因型的tc水平低于非cc基因型(p=0.028)。血脂分层后,在正常血脂亚组中,tg水平高于非cc基因型(p=0.001),hdl-c水平低于非cc基因型(p=0.041);在长寿子女组的正常血脂亚组中,cc基因型tg、hdl-c水平低于非cc基因型(p分别为0.014、0.037);在异常血脂亚组中均未发现统计学差异。bmi分层后,在正常bmi亚组内,长寿组的cc基因型的hdl-c水平低于非cc基因型(p=0.023);在平果对照组中,cc基因型的tc、ldl-c水平高于非cc基因型(p分别为0.010、0.013)。4.obfc1rs4387287基因型、年龄等因素与血脂水平的回归分析tc、tg、ldl-c水平主要与bmi、性别、年龄、舒张压呈现正相关;hdl-c水平主要与bmi、性别、年龄、体重、舒张压呈现负相关。性别分层后,在长寿子女组及当地对照组的男性人群中,OBFC1 rs4387287基因型(AA)与TC水平呈现正相关;在平果对照组男性中,OBFC1 rs4387287基因型(AA)与TC、TG水平呈现负相关。结论:1.OBFC1 rs4387287多态性与TC、TG水平相关,其A等位基因可能与平果地区人群较低的血脂水平有关。2.OBFC1 rs4387287多态性的A等位基因在平果对照组人群中有富集现象,与该地男性群体较低的TC、TG水平有关;但其对广西红水河流域人群的血脂水平影响不明显。
[Abstract]:Background: the oligonucleotide binding fold (oligonucleotide/oligosaccharide binding fold gene 1 containing 1 OBFC1) with homology with the Stn1 gene of the yeast, and is directly involved in the regulation of telomere biological function is one of the important genes of [1,2]. so far, the domestic and foreign research on the correlation between OBFC1 gene polymorphism and blood lipids is poorly understood, in order to understand the relationship between the gene and blood lipids. We carried out relevant research. Objective: To investigate the relationship between OBFC1 rs4387287 polymorphism and serum lipid levels, to understand the effect of this gene on Guangxi Red River Basin longevity population. The clinical results: 1. the data (blood lipids, blood glucose and blood pressure) between the two groups in longevity Group TG, HDL-C level is lower than the longevity of children group and local controls (all P0.05), but SBP, DBP shows the opposite trend. Pingguo control group TC, LDL-C, SBP, blood glucose level is lower than the longevity of the children group And the control group (P0.05).2.obfc1rs4387287 genotype distribution and allele frequency in the general population, the genotype CC genotype is more common, the frequency is 71.3%; the frequency of Ca genotype was 24.5%; the frequency of AA genotype was 4.2%; the frequency of allele C was 83.6%; a the allele frequency of 16.4%.a allele in the longevity Group was 16.2% in the children group; longevity frequency was 15.8% in the control group; the local frequency is 15.3% in the control group; Pingguo frequency was 19.2%, the A allele frequency in Pingguo control group than the other three groups (P0.05).
