【摘要】:1.研究目的本实验旨在从动物行为学以及性激素E2、T、PRL含量两方面对逍遥散免煎颗粒剂以及逍遥中空栓进行治疗效果的对比,探讨在中医传统理论指导下两种剂型对肝郁证大鼠的治疗作用,为临床合理使用逍遥散提供支持。2.研究方法用1月龄雌性SD大鼠50只,随机分为5组:空白对照组、肝郁模型组、逍遥散免煎颗粒剂组、逍遥中空栓高剂量组、逍遥中空栓低剂量组,每组10只,通过肝郁慢性不可预知的多种应激刺激随机组合法复制肝郁模型,21天后每组做阴道涂片连续5天,与此同时做行为学检测,逍遥中空栓高、低剂量组塞肛纳药;逍遥散免煎颗粒剂组灌胃,连续给药10天,给药结束后当日继续做阴道涂片5天,与此同时做行为学检测,结束后于第二天处死动物,取血备用。3.研究内容3.1一般状态、体重、摄食实验开始后,每天观察大鼠的一般状态,大鼠体重增长情况、摄食量每3天记录一次。3.2阴道涂片检测分别于造模后、给药后做大鼠阴道涂片检测,连续5天,用显微镜观察各大鼠动情周期是否紊乱。3.3行为学检测分别于造模后、给药后做旷场实验、高架十字迷宫实验、糖水偏嗜实验,检验造模是否成功以及给药是否有效。3.4性激素检测将腹主动脉所取的血离心,分离血清后,放射免疫分析法测定血清E2、T、PRL的含量。3.5统计学采用SPSS17.0统计软件进行分析,计量资料以均数±标准差表示,组间比较用独立样本t检验,组内比较用配对t检验,定量资料组间比较采用方差分析,P0.05为差异有统计学意义。4.结果4.1造模成功判定:与空白对照组比较,造模后肝郁模型组、逍遥散免煎颗粒剂组、逍遥中空栓高剂量组、逍遥中空栓低剂量组的一般状态、体重、摄食以及行为学的均值上观察有显著变化,经t检验分析,P0.05差异有统计学意义,提示造模成功。4.2各组阴道涂片的观察:造模后空白对照组5天涂片中动情前期、动情期、动情间期、静止期规律交替出现,仅有1天镜下可见典型角化细胞增多;肝郁模型组镜下5天中会有3-5天连续出现角化细胞,逍遥散免煎颗粒剂组、逍遥中空栓高剂量组、逍遥中空栓低剂量组给药后镜下5天中角化细胞有规律出现,部分已经恢复正常动情周期。4.3通过对行为学数据分析发现:旷场实验中,从外周格、中央格穿越格数上,逍遥散免煎颗粒剂组、逍遥中空栓高剂量组、逍遥中空栓低剂量组与肝郁模型组相比P0.05,有统计学意义;从站立次数以及理毛修饰次数上来说,逍遥中空栓高剂量组P0.05,有统计学意义;高架十字迷宫实验中,从开臂时间百分比上,逍遥散免煎颗粒剂组、逍遥中空栓高剂量组、逍遥中空栓低剂量组与肝郁模型组相比P0.05,有统计学意义;糖水偏嗜实验中,逍遥散免煎颗粒剂组、逍遥中空栓高剂量组、逍遥中空栓低剂量组糖水偏嗜度与肝郁模型组相比P0.05,有统计学意义;其余组间比较经统计学分析P0.05,认为无差异。4.4通过对性激素E2、T、PRL检测结果分析:逍遥散免煎颗粒剂组、逍遥中空栓高剂量组T、PRL与肝郁模型组相比P0.05,有统计学意义;逍遥中空栓低剂量组PRL与肝郁模型组相比P0.05,有统计学意义;其余组间比较经统计学分析P0.05,认为无差异。5.结论5.1通过对大鼠进行21天肝郁慢性不可预知的多种应激刺激随机组合法,可以成功复制出肝郁证模型。5.2肝郁模型的雌性大鼠一般状态差,体重下降,摄食减少,动情周期紊乱,旷场实验中水平和垂直得分降低,高架十字迷宫实验中开臂停留时间及次数比例下降,糖水偏嗜度降低。5.3逍遥中空栓可改善肝郁证雌性大鼠的一般状态、增加体重、摄食增多,上调旷场实验中水平和垂直得分、高架十字迷宫实验中开臂停留时间及次数比例、糖水偏嗜程度以及改善紊乱的动情周期、升高血清E2、PRL含量、降低血清T含量,与逍遥散免煎颗粒剂比较无差异,可以作为一种新剂型应用于临床。
[Abstract]:1 . The aim of this study was to compare the therapeutic effects of Xiaoyao Powder and Xiaoyao hollow suppository from the aspects of animal behavior and sex hormone E2 , T and PRL levels . The rats were randomly divided into five groups : blank control group , liver depression model group , Xiaoyao powder free decoction granule group , Xiaoyao hollow suppository high dose group and Xiaoyao hollow suppository low dose group .
