改良型磨损颗粒诱导小鼠颅骨溶解模型的构建

【摘要】:目的探讨建立一种新型、简单、经济、易操作的磨损颗粒诱导小鼠颅骨溶解模型,便于无菌性假体松动动物模型构建。方法制备无菌盐包埋磨损颗粒;实验分为3组:A组为生理盐水组;B组为纯盐组;C组为盐包埋磨损颗粒组;14 d后统一处死动物取出小鼠颅骨,进行病理学分析。结果 (1)HE染色A组和B组无明显差别,C组骨质疏松明显,骨质结构紊乱,骨吸收增加,骨膜及骨组织细胞数量明显增加,骨内细胞数量(个/视野)A组(25±3),B组(27±2),C组(45±5);骨膜炎症细胞(个/视野):A组(1 540±368)、B组(1 610±535)、C组(3 478±376)。(2)电镜下计算颅顶骨吸收陷窝面积与视野面积的百分比:A组(2.76±0.48)%、B组(2.95±0.63)%,C组(13.33±2.49)%,C组与A组、B组比较,差异有统计学意义(P0.05)。(3)TRAP染色中破骨细胞数(个/视野):A组为(3.842 7±1.182 1);B组为(4.026 1±1.372);C组(17.325±4.210 7)。A组与B组相比,无明显差别(P0.05);C组与A、B组比较:破骨细胞数量明显增加,溶骨效应增强(P0.01),TRAP染色与HE染色结果相符。结论盐溶法构建磨损颗粒诱导小鼠颅骨溶解模型是一种简单、经济、可行性强的实验方法,可以为研究人工关节无菌性松动的提供较为确切、简洁的临床实验基础,是一种值得倡导的实验方法。
[Abstract]:Objective to establish a new, simple, economical and easy to operate wear particle model of mouse skull dissolution, which is convenient for the establishment of animal model of aseptic prosthesis loosening. The experiment was divided into 3 groups: normal saline group; Group B was pure salt group; Group C was salt embedded wear particle group. After 14 days, all the animals were killed to remove the cranial bones of the mice for pathological analysis. Results there was no significant difference between group A and group B in osteoporosis, disorder of bone structure and increase of bone resorption. The number of cells in periosteum and bone tissue increased obviously, and the number of cells in bone in group A (25 卤3) and group B (27 卤2) and in group C (45 卤5); Inflammatory cells of periosteum (1 540 卤3 68%) in group A (1 610 卤5 35). The percentage of the area of resorption lacuna of the parietal bone and the area of visual field in group C was calculated by electron microscope (2.76 卤0.48%). Group B (2.95 卤0.63) and group C (13.33 卤2.49) were compared with group A and group B (P < 0.05). There was a significant difference in osteoclast number in P 0.05U. P 3 trap staining (P = 3.8427 卤1.1821) in group A (P < 0.05). The number of osteoclasts in group A was 3.842 7 卤1.182 1% (P < 0.05). Group B was 4.0261 卤1.372U; There was no significant difference between group C (17.325 卤4.210 7) and group B (P 0.05). The number of osteoclasts in group C was significantly higher than that in group A (P 0.01). Conclusion it is a simple, economical and feasible method to construct the model of mouse skull dissolution induced by wear particles by salt solution. It can provide a more precise and succinct clinical experimental basis for the study of aseptic loosening of artificial joints. It is an experimental method worthy of advocacy.
