五脏背俞穴埋线对应激衰老模型大鼠HPA轴的调节作用

【摘要】:目的衰老是由于生物体随着时间的流逝而在结构和机能上出现衰退,在适应能力和抵抗能力上出现降低的现象。由于当今生活环境不断改善,人类寿命逐步延伸,使得整个人类社会日趋老龄化,以致研究如何"抗衰老"的意义日渐凸显。本实验通过对健康大鼠注射D-半乳糖溶液合并制动应激处理,欲制造出贴近由于内外因素共同作用致衰的人类自然衰老模型,将大鼠行为学,胸腺指数以及与神经-内分泌-免疫系统(HPA轴)的相关激素作为检测指标,观察五脏背俞穴埋线对大鼠一般行为学、胸腺指数、血清皮质醇(Cor)、血浆促肾上腺皮质激素(ATCH)以及下丘脑促肾上腺皮质激素释放激素(CRH)的影响,综合阐述埋线对衰老机体的调节作用,以便为日后相关抗衰实验提供理论依据。方法SPF级,健康3月龄,雄性SD大鼠60只,体重200-250g。以体重作为分层条件,随机区组设计分为6组:空白对照组(A组)、D-半乳糖组(B组)、D-半乳糖+埋线组(C组)、D-半乳糖+应激组(D组)、D-半乳糖+应激+埋线组(E组),每组12只。每日给予B组、C组、D组和E组大鼠腹腔注射D-半乳糖溶液,A组则给予腹腔注射等量的生理盐水,共8周。D组和E组于第五周开始,给予制动应激,连续应激28日。从实验开始时即每日观察大鼠的精神状态、毛发形态,并称量记录大鼠体重。8周后,各组大鼠主动脉采血,分离血清、血浆,运用酶联免疫吸附反应法(ELISA)检测各组大鼠Cor、ATCH含量。并断头开颅,取出下丘脑,组织匀浆,取上清液,运用酶联免疫吸附反应法(ELISA)检测CRH含量。最后,取出胸腺,测量胸腺指数。运用统计学方法对比各组实验数据,采用SPSS22.0统计软件进行统计学处理。结果1.A组大鼠体重增长最多(P0.05),B组次之,C组次之,E组次之,D组最少(P值均0.05);2.A组大鼠胸腺指数最大(P0.05),B组次之,C组次之,E组次之,D组最少(P值均0.05);3.D组大鼠Cor、ATCH、CRH均处于较高范畴(P0.05),E组次之,B组次之,C组次之,A组最少(P值均0.05),差异具有统计学意义。结论1.五脏背俞穴埋线可以改善机体在衰老过程中所出现的体重减轻、精神低落、毛发枯黄等一般行为学表现。2.五脏背俞穴埋线对于机体衰老过程中所出现的胸腺萎缩状态,具有一定的改善作用。3.五脏背俞穴埋线对于机体衰老过程中所出现的HPA轴功能亢进,具有一定的抑制作用。
[Abstract]:Objective Aging is a decline in the structure and function of organisms over time and a decrease in their ability to adapt and resist. As a result of the continuous improvement of the living environment today, the life span of human beings is gradually extending, As a result of the aging of the whole human society, the significance of the study on how to "resist aging" becomes more and more prominent. This experiment was done by injecting D- galactose solution into healthy rats combined with immobilization stress. In order to create a human model of natural aging, which was induced by internal and external factors, rat behavior, thymus index and related hormones related to the neuroendocrine and immune system (HPA axis) were used as indicators. To observe the effects of embedding thread on general behavior, thymus index, serum cortisol Corn, plasma adrenocorticotropic hormone (ACTH) and hypothalamic corticotropin releasing hormone (CRH) in rats. In order to provide theoretical basis for related anti-aging experiments in the future, 60 male SD rats of SPF grade, healthy 3 months old and weight of 200-250 g were used as stratification conditions. The randomized block design was divided into 6 groups: control group A, group A, group A, group B, group B, group B, group B, group B, group C, group C, group C, group C, group C, group D, group D, group D, group D, group D, group D, group E, group E, catgut embedding with D-galactose stress, group E, 12 rats in each group, given daily to group B, with 12 rats in each group. Rats in group C, group D and group E were injected intraperitoneally with Dgalactose solution. Group A was given intraperitoneal injection of the same amount of normal saline. The rats in group D and group E were given immobilization stress at 5th weeks and continuous stress on 28th. The mental state and hair morphology of rats were observed daily at the beginning of the experiment, and the blood samples were collected from aorta of each group after weighing and recording the weight of rats. Serum and plasma were isolated, and the content of Coranach was detected by Elisa. The hypothalamus, homogenate and supernatant were removed from hypothalamus and supernatant, and the content of CRH was detected by enzyme linked immunosorbent assay (Elisa). Finally, the contents of CRH were determined by enzyme-linked immunosorbent assay (Elisa). The thymus gland was taken out, the thymus index was measured, and the experimental data of each group were compared by statistical method. Results 1. Weight gain of rats in group A was the highest in group B and group B was followed by group C, followed by group E, followed by group D and the lowest P value. 2. Group A, group A, rats with the largest thymus index, group B, group C, group C, group E, group D, second group, group D, group D, the second group. The minimum P values of group D were 0.05 and 0.05 respectively. The CRH of group D was in the higher category. Group B, group B, group C, group C, followed by group A, the minimum P values were 0.050.Conclusion 1.The embedding of the points at Wuzhangbackshu can improve the body's aging process. 2. The weight loss that occurs, General behavioral manifestation of low spirits, yellow hair, etc. 2.The state of thymus atrophy that occurs in the aging process of the body by embedding wires at the point of the back of the five viscera, It can improve the function of HPA axis and inhibit the function of HPA axis in the aging process. 3. The buried thread at Wuzangbushu point can inhibit the function of HPA axis in the process of aging.
