舒心汤对抑郁模型大鼠治疗作用的实验研究

【摘要】:目的通过舒心汤干预孤养与慢性温和不可预见性应激(CUMS)抑郁模型大鼠的行为学及下丘脑-垂体-肾上腺轴(HPAA)和炎性因子白介素1β(IL-1β)、白介素6(IL-6)的结果差异,观察舒心汤对抑郁模型大鼠的疗效。根据中医调理脏气理论,探讨舒心汤的抗抑郁机制,为临床应用舒心汤治疗抑郁症提供新的思路。方法以孤养结合CUMS方法制造抑郁大鼠模型,分别于实验前和实验后测定大鼠的体重、糖水消耗率、敞箱实验得分、强迫游泳不动时间的变化结果,其中体重情况每周测定一次,据此评定舒心汤对孤养结合CUMS抑郁模型大鼠行为学的改变情况。按照ELISA试剂盒说明书分别测量血清促肾上腺皮质激素(ACTH)、皮质醇(CORT)、白介素6(IL-6)、白介素-1β(IL-1β)水平。RT-PCR检测下丘脑内CRHm RNA及海马内GRm RNA的表达。实验数据以(?)±s表示,组间比较应用单因素方差分析进行统计,以P0.05为有差异性,P0.01为有显著差异。结果1 28天的造模应激之后,与空白组对比,模型组的行为学指标均明显降低(P0.05或P0.01),模型组大鼠的血清ACTH、CORT、IL-1β、IL-6及下丘脑CRHm RNA水平明显升高(P0.05或P0.01),海马GRm RNA表达明显下降(P0.05或P0.01),结果有(明显)差异性。2 28天药物干预之后,与模型组对比,舒心汤组、舍曲林组及联合用药组的行为学指标明显上升(P0.05或P0.01),血清ACTH、CORT、IL-1β、IL-6及下丘脑CRHm RNA水平明显下降(P0.05或P0.01),海马GRm RNA表达明显上升(P0.05或P0.01),结果有(明显)差异性。结论1中药复方制剂舒心汤能有效改善孤养结合CUMS抑郁模型大鼠的行为学指标,即加速体重的增长,增加糖水消耗率,增加敞箱实验垂直和水平运动得分,减少强迫游泳大鼠不动时间。2中药复方制剂舒心汤能调节抑郁模型大鼠的HPA轴及炎性因子功能,即降低抑郁大鼠血清中ACTH、CORT、IL-1β、IL-6的水平及抑郁大鼠下丘脑CRH的基因表达,提高抑郁大鼠海马GR的基因表达。3舒心汤的抗抑郁机制可能与干预神经内分泌免疫系统的HPA轴及炎性因子有关。
[Abstract]:Objective to study the behavior and the difference of hypothalamus-pituitary-adrenal axis (HPAA) and inflammatory factor IL-1 尾 (IL-1 尾, interleukin-6) in rats with depression induced by Shuxin decoction and chronic mild and unpredictable stress (CUMS).To observe the effect of Shuxin decoction on depression model rats.According to the theory of regulating visceral qi in TCM, the antidepressant mechanism of Shuxin decoction is discussed, which provides a new idea for clinical application of Shuxin decoction in the treatment of depression.Methods the depressive rat model was established by the method of solitary raising and CUMS. The weight, the consumption rate of sugar water, the score of open box test and the time of forced swimming were measured before and after the experiment, in which the body weight was measured once a week.To evaluate the effect of Shuxin decoction on the behavior of rats with solitary care combined with CUMS depression.The expression of CRHm RNA in hypothalamus and GRm RNA in hippocampus were detected by reverse transcription-polymerase chain reaction (RT-PCR) according to the instructions of ELISA kit.The experimental data were expressed as) 卤s, and the statistical results were analyzed by univariate analysis of variance (ANOVA). There was significant difference between the two groups with P05 as the difference (P0.01).Results after 28 days of stress, the model was compared with the control group.The behavioral indexes of the model group were significantly lower than those of the control group (P0.05 or P0.01). The serum levels of IL-1 尾 -IL-6 and CRHm RNA in the hypothalamus of rats in the model group were significantly higher than those in the control group (P 0.05 or P 0.01), and the expression of GRm RNA in the hippocampus decreased significantly (P 0.05 or P 0.01). The results showed that there were significant differences between the two groups after the drug intervention for 2.28 days.Compared with the model group, the behavioral indexes of Shuxin decoction group, sertraline group and combination group were significantly increased (P 0.05 or P 0.01), the serum levels of ACTH cortisone IL-1 尾 IL-6 and hypothalamus CRHm RNA were significantly decreased (P0.05 or P0.01), and the expression of GRm RNA in hippocampus was significantly increased (P 0.05 or P 0.01).Conclusion (1) Shuxin decoction, a compound preparation of Chinese medicine, can effectively improve the behavioral indexes of rats with solitary rearing combined with CUMS depression model, that is, accelerate the growth of body weight, increase the consumption rate of sugar water, and increase the scores of vertical and horizontal movement in open box experiment.To reduce the immobility time of forced swimming rats. 2. Shuxin decoction can regulate the function of HPA axis and inflammatory factors in depressive rats, that is, it can reduce the level of serum ACTHCORTT IL-1 尾 and IL-6 and the gene expression of CRH in hypothalamus of depressive rats.The antidepressant mechanism of Shuxin decoction may be related to the intervention of HPA axis and inflammatory factors in neuroendocrine immune system.
