慢性间断性低氧对大鼠海马神经元及生长相关蛋白的进行性影响

【摘要】:目的观察慢性间断性低氧(CIH)过程中大鼠海马神经元的组织结构及生长相关蛋白(GAP)-43表达的进行性变化。方法成年雄性Sprague-Dawley大鼠40只,随机分为对照组、CIH 1周组、CIH 3周组和CIH 5周组,每组各10只。后三组大鼠在自制缺氧舱内每天缺氧8 h,连续缺氧1周、3周和5周。HE染色后在光学显微镜下观察海马神经元组织结构;免疫组化方法检测海马神经元GAP-43的表达。结果 CIH大鼠海马出现变性坏死神经元,并且随着缺氧时间延长,上述改变呈逐渐加重趋势。与对照组比较,CIH组大鼠海马神经元GAP-43阳性表达增多,CIH后1周出现表达高峰(P=0.038),CIH后3周表达逐渐减少(P=0.382),CIH后5周表达接近正常水平(P=0.860)。结论随着缺氧时间延长,变性坏死神经元数量呈进行性增多,而GAP-43的表达却与此不一致。
[Abstract]:Aim to observe the progressive changes in the structure of hippocampal neurons and the expression of growth associated protein (GAP-43) in rat hippocampal neurons during chronic intermittent hypoxia (CIH). Methods Forty adult male Sprague-Dawley rats were randomly divided into two groups: the control group (n = 10) and the control group (n = 10). The structure of hippocampal neurons was observed under optical microscope after hypoxia for 8 hours per day in the last three groups, and the expression of GAP-43 in hippocampal neurons was detected by immunohistochemical method. Results denatured and necrotic neurons appeared in the hippocampus of CIH rats. Compared with the control group, the expression of GAP-43 in hippocampal neurons in CIH group was increased. The expression of GAP-43 in hippocampal neurons of CIH group was decreased gradually at 3 weeks after CIH, and the expression of GAP-43 was close to the normal level at 5 weeks after CIH. Conclusion with the prolongation of hypoxia time, the number of denatured and necrotic neurons increased progressively, but the expression of GAP-43 was not consistent with this.
