苏州市甲型H1N1流感病毒神经氨酸酶基因的分子演变和遗传特征分析

【摘要】:目的探讨新型H1N1流感病毒从2009年在苏州出现至2011年基本消失的整个过程中,苏州分离株的神经氨酸酶(Neuraminidase,NA)的分子特征及其基因变异和发展规律。方法提取苏州分离株的RNA,以WHO推荐的引物扩增NA基因并测序。以WHO推荐的季节性H1N1和新型H1N1疫苗株为参照,采用生物信息学软件对苏州毒株的NA进行详细的分子特征分析,并与全球及全国的代表性毒株进行序列进化分析。结果与疫苗株相比,5株苏州株的核苷酸序列同源性均值为99.64%,氨基酸序列同源性均值为99.36%。5株苏州株中都没有发现抗原位点的改变,但4株在耐药性相关位点附近出现N248D突变,还有1株2011年株增加了1个新的潜在糖基化位点。结论首次详细分析了苏州地区2009年以来H1N1型流感病毒NA的分子特征,表明苏州毒株出现了一个新的潜在糖基化位点,但抗原性和药物敏感性均保持稳定。
[Abstract]:Objective to investigate the molecular characteristics, gene variation and development of neuraminidase (na) of the new H1N1 influenza virus in Suzhou from 2009 to 2011. Methods RNA was extracted from Suzhou isolate. Na gene was amplified and sequenced with primers recommended by WHO. Taking the seasonal H1N1 and new H1N1 vaccine strains recommended by WHO as reference, the molecular characteristics of na of Suzhou strain were analyzed by bioinformatics software, and the sequence evolution analysis was carried out with the representative strains in the world and the whole country. Results the average nucleotide sequence homology and amino acid sequence homology were 99.64 and 99.36.5, respectively, compared with the vaccine strains. However, N248D mutations were found in 4 strains near drug-resistance related sites. Another 2011 strain added a new potential glycosylation site. Conclusion the molecular characteristics of H1N1 influenza virus na in Suzhou region since 2009 were analyzed in detail for the first time. The results showed that there was a new potential glycosylation site in Suzhou strain, but the antigenicity and drug sensitivity remained stable.
