间日疟原虫环子孢子蛋白PCR-RFLP多态性分析

【摘要】:采集2011年河南省间日疟原虫阳性患者血样,用限制性片段长度多态性聚合酶链反应(PCR-RFLP)法分析环子孢子蛋白(circumsporozoite protein,CSP)基因型,并结合流行病学资料进行初步种群分布分析。共采集间日疟患者血样37份,根据流行病学调查确定河南当地感染病例血样26份,输入性病例血样11份。37份血样均扩增出1条约750 bp的条带。PCR-RFLP结果显示,CSP基因PV-Ⅱ型1份,占2.7%,PV-Ⅰ型36份,占97.3%。26份当地病例血样CSP基因均为PV-Ⅰ型,其中PV-Ⅰ-a型占42.3%(11/26),PV-Ⅰ-b型占57.7%(15/26);11份输入病例血样中,PV-Ⅰ-a型占90.9%(10/11),PV-Ⅱ型占9.1%(1/11),未见PV-Ⅰ-b型。提示河南省当地流行的间日疟病例中,CSP基因PV-Ⅰ-b型略多于PV-Ⅰ-a型;输入性间日疟病例中PV-Ⅰ-a型占优势。
[Abstract]:Blood samples of Plasmodium vivax positive patients in Henan Province were collected in 2011. The genotypes of circumsporozoite protein were analyzed by restriction fragment length polymorphism polymerase chain reaction (PCR-RFLP), and the preliminary population distribution was analyzed with epidemiological data. A total of 37 blood samples were collected from vivax malaria patients. 26 blood samples were collected from local infection cases in Henan Province, and 11 blood samples from imported cases were collected.
