激光致猴脉络膜新生血管模型的应用

【摘要】:目的通过激光诱导猴眼底黄斑区脉络膜新生血管(CNV)渗漏和生长,建立与人脉络膜新生血管类似的动物模型,对拟用于治疗脉络膜新生血管相关疾病药物的药效学进行评价。方法采用眼科激光光凝仪围绕猴黄斑中心凹进行光凝造模,光凝后2~3周,进行荧光素眼底血管造影检查,筛选出造模成功的动物,根据造模效果分为模型对照组、阳性对照组、药物低、中、高剂量组。分别于给药前、给药后14和28 d进行眼底彩色照相、荧光素眼底血管造影(FFA)、光学相干断层扫描(OCT)检查,给药后29 d解剖动物进行眼球组织病理学检查。结果阳性对照组、药物低、中、高剂量组给药后荧光素渗漏面积明显减少,激光斑病损处视网膜厚度明显降低。组织学切片观察到模型对照组CNV视网膜神经上皮脱离及神经上皮下水肿,阳性对照组、药物低、中、高剂量组眼球基本未见异常组织学改变。结论该药物在各剂量下均对激光所致的猴脉络膜新生血管具有较明显的抑制作用。激光诱导的猴脉络膜新生血管模型稳定持久,是研究治疗与脉络膜新生血管相关眼底病药物的理想动物模型。
[Abstract]:Objective to establish an animal model of choroidal neovascularization similar to human choroidal neovascularization by laser induced choroidal neovascularization (CNV) leakage and growth in the macular region of monkey eye, and to evaluate the pharmacodynamics of the drugs proposed for the treatment of choroidal neovascularization related diseases. Methods the ophthalmic laser photocoagulation instrument was used to model the macular fovea of monkey. After 2 weeks of photocoagulation, fundus fluorescein angiography was performed. The successful animals were selected and divided into model control group according to the model effect. Positive control group, low, medium and high dose group. Fundus color photography was performed before administration, 14 d and 28 d after administration, fundus fluorescein angiography (FFA),) optical coherence tomography (OCT) and histopathological examination of eyeball were performed 29 days after administration. Results in the positive control group, the area of fluorescein leakage was significantly decreased and the retinal thickness of laser spot lesion was significantly decreased in the low, middle and high dose groups. Histopathological examination showed that the retinal neuroepithelial detachment and subcutaneous edema of CNV were observed in the model control group, but no abnormal histological changes were observed in the low, medium and high dose groups of the positive control group. Conclusion the drug has obvious inhibitory effect on the choroidal neovascularization induced by laser at various doses. The laser-induced choroidal neovascularization model is an ideal animal model for the treatment of choroidal angiogenic fundus disease.
