双黄消痤丸对兔耳痤疮模型毛囊面积、皮脂腺的影响

【摘要】:目的:通过双黄消痤丸对兔耳痤疮模型毛囊面积、皮脂腺数量和皮脂腺直径影响的研究,探讨双黄消痤丸对机体抗角化和改善皮脂腺的作用机理,为临床治疗痤疮提供实验依据。方法:将48只12周龄的新西兰雄性大白兔随机分为6组:双黄消痤丸高、中、低剂量组、阳性对照组、模型对照组、空白对照组,每组各8只。除空白对照组以外,其余各组均建立兔耳痤疮模型。造模成功后,高、中、低剂量组各给药量分别为1.68g/kg、0.84g/kg、0.42g/kg,阳性对照组(丹参酮胶囊组)给药量为0.187g/kg,空白组与模型组给予同体积的0.9%氯化钠溶液,日一次,连续14天。于末次给药20h后,在兔的耳缘静脉中注射20ml的空气,将其处死。然后迅速用1cm打孔器在右耳标记处取皮肤组织块,并用10%福尔马林进行固定,石蜡包埋,切片,HE染色,于光镜下观察其组织形态学的改变,记录毛囊面积、皮脂腺数量和皮脂腺直径。结果:1、双黄消痤丸各组与丹参酮胶囊对照组均能缩小兔耳痤疮模型皮脂腺的直径,且双黄消痤丸高剂量组疗效优于低剂组(P0.01);2、双黄消痤丸高剂组能使痤疮模型中正常的皮脂腺数量明显增多,使正常结构的皮脂腺数量趋于正常水平(P0.05);3、双黄消痤丸高剂量组对痤疮模型毛囊面积影响显著,能显著缩小增生的毛囊面积(P0.01)。结论:双黄消痤丸能减少实验兔耳皮脂腺数量、缩小皮脂腺的直径及毛囊面积,对调节皮脂腺分泌量、缩小毛囊、抗角化等方面的效果显著。
[Abstract]:Objective: to study the effects of Shuanghuangxiaocuo pills on hair follicle area, sebaceous gland quantity and sebaceous gland diameter in rabbit ear acne model, and to explore the mechanism of Shuanghuangxiaocuo pill on anti-keratosis and improvement of sebaceous gland. To provide experimental basis for clinical treatment of acne. Methods: 48 New Zealand male rabbits aged 12 weeks were randomly divided into 6 groups: Shuanghuangxiaocuo pills high, middle and low dose groups, positive control group, model control group and blank control group, each group had 8 rabbits. Except the blank control group, all the other groups established the rabbit ear acne model. After the establishment of the model, the dosage of high, medium and low dose groups were 1.68 g / kg / kg 0.84 g / kg and 0.187 g / kg, respectively. The control group (tanshinone capsule group) was given 0.187 g / kg of sodium chloride solution, and the blank group and model group were given 0.9% sodium chloride solution in the same volume once a day for 14 days. After the last administration for 20 hours, 20ml was injected into the auricular vein of rabbits and killed. Then the skin tissue mass was quickly taken from the right ear label with 1cm perforator, fixed with 10% formalin, embedded in paraffin, stained with HE. The histomorphology of the tissue was observed under light microscope, and the hair follicle area was recorded. The number of sebaceous glands and the diameter of sebaceous glands. Results: the diameter of sebaceous gland of acne model was reduced in each group of Shuanghuangxiaocuo pill group and tanshinone capsule control group. The therapeutic effect of Shuanghuangxiaocuo pills in high dose group was better than that in low dose group (P0.01), and Shuanghuangxiaocuo pills high dose group could significantly increase the number of normal sebaceous glands in acne model. The number of sebaceous glands of normal structure tended to normal level (P0.05). The high dose group of Shuanghuangxiaocuo pills had a significant effect on the hair follicle area of acne model and could significantly reduce the accretive hair follicle area (P0.01). Conclusion: Shuanghuangxiaocuo pills can reduce the number of sebaceous glands, reduce the diameter of sebaceous glands and the area of hair follicles, regulate the secretion of sebaceous glands, reduce hair follicles and resist keratosis.
