2011~2012年武汉地区肠道病毒71型VP1基因的遗传进化分析

【摘要】:目的观察武汉地区手足口病(HFMD)患儿肠道病毒71型(EV71)的基因型及其重组特点。方法收集2011年4月至2012年3月武汉地区1 844例HFMD患儿EV71阳性的咽拭子标本并进行病毒分离鉴定,对124株EV71病毒分离株的508 bp VP1基因序列进行测序,选择2株来自重症和1株来自轻症患儿的EV71毒株,构建VP1序列系统进化树并分析其全基因组序列的重组特点。结果 VP1序列系统进化分析显示,124株EV71型均属于C4基因型,其中56株属于C4a亚群,68株属于C4b亚群,Bootscan和5'UTR、P1、P2、P3区的进化树证实3株EV71分离株均为亲本株C4a型毒株guangdong/GD/CHN/2009和C4b型毒株SHZH98/GD/CHN/1998在P2区2A-2B基因交界处发生型内重组所产生。结论 2011年4月至2012年3月武汉市及周边地区的EV71分离株均为C4基因型,与1998年以来中国大陆的优势株流行一致,且C4基因型存在型内重组现象。
[Abstract]:Objective to observe the genotype and recombinant characteristics of enterovirus 71 (EV71) in children with hand-foot-mouth disease (HFMD) in Wuhan. Methods from April 2011 to March 2012, EV71 positive throat swabs of 1 844 children with HFMD were collected and identified. The 508 bp VP1 gene sequences of 124 EV71 virus isolates were sequenced. Two EV71 strains from severe disease and one from mild disease were selected to construct the phylogenetic tree of VP1 sequence and analyze the recombination characteristics of the whole genome sequence. Results the phylogenetic analysis of VP1 sequence showed that the EV71 genotype of 124strain belonged to C4 genotype. Among them, 56 strains belong to C4a subgroup, 68 strains belong to C4b subgroup Bootscan and 5UTRP1P2P2P3 region. It is confirmed that the three isolates of EV71 were all generated by recombination of parental strain C4a strain guangdong/GD/CHN/2009 and C4b strain SHZH98/GD/CHN/1998 at the junction of 2A-2B gene in P2 region. Conclusion from April 2011 to March 2012, EV71 isolates from Wuhan and its surrounding areas were all C4 genotypes, which were consistent with the prevalence of dominant strains in mainland China since 1998.
