HSP90β在小鼠生殖系统中的表达及调控

【摘要】:目的研究热休克蛋白90β(HSP90β)在小鼠睾丸、附睾、精子中的表达及调控机制。方法从mRNA和蛋白水平上对HSP90β在小鼠睾丸和附睾组织细胞中及精子上表达及定位进行分析,同时对其基因在雄性生殖中的表达调控进行初步探索。结果 HSP90βmRNA在小鼠附睾中的表达量比在睾丸中高。在小鼠精子中定位于顶体部位。其基因在去势小鼠的附睾中表达降低,补充睾酮后又恢复正常,在发育中小鼠的睾丸、附睾中表达也有变化。结论 HSP90β在小鼠精子形成及受精中可能发挥重要作用;其在去势小鼠附睾中和在出生后不同阶段小鼠睾丸、附睾中的表达模式,说明其基因表达受激素调控和发育调控的影响。
[Abstract]:Objective to study the expression and regulatory mechanism of heat shock protein 90 尾 (HSP90 尾) in testis, epididymis and sperm of mice. Methods the expression and localization of HSP90 尾 in mouse testis and epididymis tissues and spermatozoa were analyzed at the level of mRNA and protein, and the regulation of HSP90 尾 gene expression in male reproduction was explored. Results the expression of HSP90 尾 mRNA in mouse epididymis was higher than that in testis. It was located in acrosome in mouse spermatozoa. Its gene expression decreased in the epididymis of ovariectomized mice, then returned to normal after testosterone supplementation, and also changed in the testis and epididymis of developing mice. Conclusion HSP90 尾 may play an important role in mouse spermatogenesis and fertilization, and its expression patterns in the testis and epididymis of ovariectomized mice and postnatal mice at different stages indicate that its gene expression is affected by hormone regulation and developmental regulation.
