人卵母细胞形态学指标预测受精和胚胎发育潜能

【摘要】:目的:探讨卵母细胞浆内单精子注射(intracytoplasmic sperm injection,ICSI)周期中人卵母细胞形态学测量的指标与受精和胚胎发育潜能之间的关系。方法:利用OCTAX Eye-ware software系统在对卵母细胞行ICSI前,逐一采集卵母细胞图像,测量透明带厚度、卵母细胞大小、极体位置的卵间隙大小和极体形态(完整或破碎),将ICSI后的卵母细胞和受精后的胚胎进行单孔培养,记录每个卵母细胞的受精情况、胚胎发育情况。对卵母细胞的形态数据与受精情况、胚胎发育情况进行逐步多元因素Logistic回归分析。结果:共测量卵母细胞436枚,受精率84.9%(370/436),胚胎可用率60.8%(225/370),卵母细胞形态学指标中,透明带厚度、极体位置的卵间隙大小和卵母细胞受精与否有关,受精卵较未受精卵有较厚的透明带[(18.0±2.3)μm vs.(16.9±2.7)μm]和较大的卵间隙[(14.4±3.3)μm vs.(13.2±3.9)μm];而卵母细胞的大小和极体形态均与取卵后72 h的胚胎发育质量有关,发育为可用胚胎的卵母细胞较发育为废用胚胎者有较大的直径[(116.6±3.7)μm vs.(114.7±3.6)μm]和较高的极体完整率(86.2%vs.66.7%)。结论:人卵母细胞形态学测量的指标可预示卵子受精和胚胎发育的潜能。
[Abstract]:Objective: To study the relationship between the index of human oocyte morphology and the potential of fertilization and embryo development in the period of intracytoplasmic sperm injection (ICSI). Methods: Oocyte images were collected one by one, and the thickness of the zona pellucida, the size of the oocytes, the size of the oocytes and the morphology of the oocytes (intact or broken) were measured by using the OCTAX Eye-ware software system. And carrying out single-hole culture on the oocytes after ICSI and the embryos after fertilization, and recording the fertilization condition and the embryo development condition of each oocyte. The morphological data of the oocytes and the conditions of fertilization and the development of the embryos were analyzed by logistic regression. Results:436 of the oocytes were measured, 84.9% (370/436), 60.8% (225/370) of the embryo availability, the thickness of the zona pellucida, the size of the zona pellucida and the fertilization of the oocytes. The fertilized egg has a thicker zona pellucida[(18.0-2.3). mu.m vs. (16.9-2.7). mu.m] and a larger egg gap[(14.4-3.3). mu.m] and a larger egg gap[(14.4-3.3). mu.m]; and the size and the morphology of the oocyte are all related to the embryo development quality of 72 hours after the oocyte retrieval, Oocytes developed to available embryos were more developed to have a larger diameter[(116.6-3.7). mu.m vs. (114.7[3.6). mu.m] and higher in the complete rate of the embryos (86.2% vs. 66.7%). Conclusion: The index of the morphology of the human oocytes may indicate the potential of the fertilization and the development of the embryo.
