海马TLR4及TNF-α参与CCI大鼠神经痛的初步研究

目的:1.阐明海马TLR4(Toll like receptor 4,Toll样受体4)及TNF-α(Tumor Necrosis Factor-alpha,肿瘤坏死因子-α)表达是否参与CCI(chronic constriction injury of sciatic nerve,坐骨神经慢性缩窄性损伤)大鼠神经痛。2.阐明双侧海马注射米诺环素或miRNA-181c激动剂能否通过下调海马TLR4及TNF-α表达缓解CCI大鼠神经痛。方法:选用56周龄SD雄性大鼠,均行海马CA1区置管。1.SD大鼠,随机分成3组(n=8):(1)假手术组(Sham组)、(2)CCI+溶剂对照组(CCI+0.01 mol/L PBS)、(3)CCI+米诺环素组(CCI+2μg/μL米诺环素)。CCI+米诺环素组双侧海马CA1区注射米诺环素(2μg/μL,2次/日,持续7 d)。假手术组及CCI+溶剂对照组双侧海马CA1区注射等体积的0.01M PBS(持续7 d)。于术前1 d,术后第1、3、5、7 d测定给药1 h后的MWT(mechanical withdraw thresh...
