丁苯酞改善血管性痴呆大鼠认知功能及其对海马区GAP-43和突触素表达的影响

【摘要】:目的探讨丁苯酞对血管性痴呆(VD)大鼠认知功能及对海马区GAP-43和突触素P38表达的影响。方法选取雄性Wistar大鼠90只,随机分为观察组、对照组、空白组各30只。空白组不采取任何操作;对照组大鼠制成VD模型,并予以生理盐水;观察组大鼠制成VD模型后,予以丁苯肽注射液。治疗1个月后,采取MG-Y-型迷宫检测其认知功能以及采用Western印迹和RT-PCR分别检测大鼠海马区内GAP-43和突触素P38的表达。结果观察组和空白组大鼠的错误反应次数均明显低于对照组(P均0.05),且空白组大鼠降低更明显(P0.05)。对照组和观察组的GAP-43在大鼠海马区内的mRNA中表达较空白组高(P均0.05),且观察组表达程度较对照组更高(P0.05);观察组的突触素P38在大鼠海马区内的mRNA中表达较空白组和对照组均提高(P均0.05),而对照组和空白组之间表达无差异(P0.05);观察组GAP-43在大鼠海马区内的蛋白中表达较对照组和空白组均提高(P均0.05),对照组和空白组之间表达无差异(P0.05);对照组和观察组的突触素P38在大鼠海马区内的蛋白中表达较空白组均提高(P均0.05),且观察组表达程度较对照组更高(P0.05)。结论对于VD大鼠,可予以丁苯酞注射液治疗,其可促进神经元的修复及再生,改善VD大鼠海马区GAP-43及突触素P38的表达,从而提供其认知功能,改善其预后。
[Abstract]:Objective to investigate the butylphthalide on vascular dementia (VD) cognitive function of rats and its effect on the expression of GAP-43 in hippocampus and synaptophysin P38. Methods 90 male Wistar rats were selected and randomly divided into the observation group, the control group and the blank group of 30 rats. The blank group did not take any operation; the rats in the control group made the VD model and given the physiological saline; the rats of the observation group were made into the VD model and gave the butadiepin injection. After 1 months of treatment, the expression by MG-Y- maze test and the cognitive function by Western blot and RT-PCR were detected in rat hippocampal area GAP-43 and synaptophysin P38. Results the number of erroneous responses in the observation group and the blank group was significantly lower than that in the control group (P 0.05), and the decrease in the blank group was more obvious (P0.05). The observation group and the control group of GAP-43 mRNA in hippocampus in rats expressed higher than those in control group (P 0.05), and the expression level of the observation group higher than the control group (P0.05); the observation group of synapsin P38 expression compared with control group and control group in the mRNA hippocampus in rats were increased (P 0.05), and between the control group and the blank group was no significant difference (P0.05); the observation group in GAP-43 protein in hippocampus of rats in the expression were higher than those in the control group and the control group (P 0.05), between the control group and the blank group was no significant difference (P0.05); the control group and the observation the P38 group than the control group significantly increased synaptophysin expression in hippocampus in protein in rats (P 0.05), and the expression level of the observation group higher than the control group (P0.05). Conclusion the VD rats can be butylphthalide injection therapy, which can promote the repair and regeneration of neurons, improve the expression of VD in rat GAP-43 and synaptophysin P38 in hippocampus, thus providing the cognitive function, improve the prognosis.

【正文快照】: 血管性痴呆(VD)的发病率近年来持续上升,现已成为中老年人常见病之一,在我国其患病率已达2.1%~3.5%〔1,2〕。而目前针对VD的治疗方法主要是通过促进脑部微循环,改善血液灌注等对症治疗。丁苯酞注射液可以改善脑卒中患者的中枢神经系统损伤以及脑能量代谢和脑缺血区的微循环。
