【摘要】：目的探讨人群中髓过氧化物酶基因-63GA和-638CA多态性与血管性痴呆相关性。方法年龄、性别方面相匹配的271例血管性痴呆患者和211例健康体检者,应用等位基因特异引物PCR (ASP-PCR)技术,检测两组被检者MPO基因-463GA和-638CA多态位点的基因型和等位基因的分布频率。采用Hardy-Weinberg平衡法检验样本基因型的吻合度,SPSS18.0统计软件进行统计学分析。结果 1研究样本人群的MPO基因-463GA和-638CA具有多态性。MPO基因-463GA多态位点的等位基因分布频率为,正常对照组：G为0.762,A为0.238,血管性痴呆组：G为0.779,A为0.221, MPO基因-638CA的等位基因频率为,正常对照组：C为0.846,A为0.154,血管性痴呆组：C为0.812,A为0.188,两组比较没有显著性差异(P0.05)。Logistic回归分析显示,-463GA和-638CA多态位点的基因型对血管性痴呆患病风险没有影响(P0.05)。 2血管性痴呆组的“-463GG”型亚组中,携带'-638CA+AA"基因型受试者,高于“-638CC”基因型携带者的糖尿病患病率(P0.05)；而‘'-463GA+AA"型亚组中,两者糖尿病的发病率比较,差异无显著性(P0.05)。血管性痴呆组的-463位点两不同基因型亚组中,-638位点不同基因型的高血压患病率没有明显差异(／0.05)。结论 1MPO基因-463GA和-638CA多态性,与血管性痴呆的发生风险无明显相关性。 2MPO基因-463GA与-638CA多态性,一定程度上影响血管性痴呆患者的患糖尿病风险。
[Abstract]:Objective To explore the correlation between the polymorphism of myeloperoxidase gene - 63GA and -638CA in the population and vascular dementia . Methods The distribution frequencies of the genotypes and alleles of MPO gene - 463GA and -638CA polymorphic loci in two groups were tested by using allele specific primer PCR ( ASP - PCR ) technique . Results The allele frequencies of MPO gene - 463GA and -638CA in the sample population were polymorphic . The allele frequencies of MPO gene - 463GA polymorphic loci were 0.762 , 0.238 , 0.779 , 0.221 , 0.154 , 0.812 , 0.188 respectively . There was no significant difference between the two groups ( P0.05 ) . Logistic regression analysis showed that the genotype of -463GA and -638CA polymorphic loci had no effect on the risk of vascular dementia ( P0.05 ) . In the " -463GG " subgroup of the vascular dementia group , the prevalence of diabetes mellitus ( P 0.05 ) was higher than that in the " -638CC " type subgroup , whereas in the ' - 463GA + AA " subgroup , there was no significant difference in the incidence of diabetes between the two groups ( P0.05 ) . There was no significant difference in the prevalence of hypertension among the two different genotypes of the -463 locus in the vascular dementia group ( P 0.05 ) . Conclusion The 1MPO gene - 463GA and -638CA polymorphism were not related to the risk of vascular dementia . 2MPO gene - 463GA and -638CA polymorphism , to some extent , affect the risk of diabetes mellitus in patients with vascular dementia .
