NSE在阿尔茨海默病模型大鼠脑组织中的蛋白表达变化

【摘要】:目的探讨阿尔茨海默病(AD)模型大鼠脑组织中的蛋白表达变化。方法实验选取Wistar雄性成年大鼠20只,随机分为正常组及AD模型组。切断大鼠双侧海马制备AD动物模型。肽指纹图谱是通过NCBI对特定差异蛋白点进行生物质谱分析、鉴定。对不同实验组海马蛋白进行测定。通过蛋白变化的比较,选取有差异的蛋白点条件:AV Ratio1.3或1.3,t检验P0.05,为有意义蛋白点。结果进一步通过SwissPort蛋白数据库进行鉴定。结果质谱分析发现AD模型组海马内有一表达明显升高的蛋白点,经鉴定为神经元烯醇化酶(NSE)。结论 NSE在AD的病程中,表达发生变化,是在AD疾病中起重要作用的酶。
[Abstract]:Objective to investigate the changes of protein expression in the brain of Alzheimer's disease (AD) rats. Methods Twenty adult Wistar male rats were selected. The animal model of AD was made by cutting off the bilateral hippocampus of rats. The peptide fingerprint was used to analyze the specific differential protein sites by NCBI. Identification. Determination of hippocampal proteins in different experimental groups. Different protein point conditions were selected, such as: AV Ratio1.3 or 1.3t test (P0.05). The results were further identified by SwissPort protein database. Results A significantly increased protein expression was found in the hippocampus of AD model group by mass spectrometry analysis. Conclusion the expression of NSE changes in the course of AD, and it is an important enzyme in AD disease.
