鹰嘴豆芽素A对过氧化氢诱导大鼠原代海马神经元损伤的作用及抗凋亡机制

【摘要】:目的探讨植物雌激素鹰嘴豆芽素A对H_2O_2诱导大鼠原代海马神经元细胞凋亡的保护作用。方法取新生24 h内的海马神经元细胞,培养10 d左右,鉴定神经细胞元纯度。用雌激素和低、中、高剂量的鹰嘴豆芽素A处理细胞24 h,除正常组外,其余组给予H_2O_2作用12 h,CCK-8法测定细胞活力,用流式细胞仪测定各组细胞凋亡率。Western blot测定Bcl-2、Bax、caspase-3蛋白表达水平。结果用神经元特异性抗体微管相关蛋白2荧光检测海马神经元纯度达98%以上,鹰嘴豆芽素A预处理24 h能显著改善H_2O_2作用12 h后的海马神经元活力。用流式细胞仪观察H_2O_2损伤后,凋亡细胞数和坏死细胞数明显增加,给予鹰嘴豆芽素A预处理后,可明显减少H_2O_2引起的凋亡和坏死细胞数。鹰嘴豆芽素A各剂量组剂量依赖性地提高模型细胞Bcl-2蛋白表达,降低Bax、caspase-3蛋白表达。结论鹰嘴豆芽素A对H_2O_2诱导大鼠原代海马神经元损伤的保护作用可能与其抑制细胞凋亡有关,鹰嘴豆芽素A抗神经元凋亡作用的机制可能与其上调Bcl-2蛋白表达和下调Bax、caspase-3蛋白表达有关。
[Abstract]:Objective to investigate the protective effect of phytoestrogenin A on apoptosis of primary hippocampal neurons induced by H _ 2O _ 2 in rats. In addition to the normal group, the other groups were treated with H _ 2O _ 2 for 12 h to determine the cell viability by CCK-8 method. Apoptosis rate was measured by flow cytometry. Western blot was used to detect the expression of Bcl-2Baxax-caspase-3 protein. Results the purity of hippocampal neurons was more than 98% by fluorescence detection of neuron specific antibody microtubule-associated protein 2. The hippocampal neuronal activity of H2O2 was significantly improved after H2O2 was pretreated for 24 h. The number of apoptotic cells and necrotic cells increased significantly after H _ 2O _ 2 injury was observed by flow cytometry, and the number of apoptotic cells and necrotic cells were observed by flow cytometry. The number of apoptotic and necrotic cells induced by H _ 2O _ 2 was significantly reduced. The expression of Bcl-2 protein in model cells was increased in a dose-dependent manner. Conclusion the protective effect of chickpein A on the injury of primary hippocampal neurons induced by S _ 2O _ 2 in rats may be related to its inhibition of apoptosis. The mechanism of antiapoptotic effect of chickpein A may be related to its up-regulation of Bcl-2 protein expression and down-regulation of Bax-caspase-3 protein expression.
