急性创伤应激对小鼠行为变化的影响

【摘要】:目的研究急性创伤应激对小鼠自发行为、焦虑、学习记忆的影响。方法将16只小鼠分为实验组(n=8)和对照组(n=8),实验组给予足底电击,对照组不给予任何处理;分别在处理后0 h、12 h、7 d进行旷场实验以及高架十字迷宫实验,以评价小鼠行为学改变;用水迷宫实验以评价小鼠学习记忆改变。结果 (1)两组小鼠的站立次数和中央区域活动时间、路程在应激后0 h和12 h差异均有统计学意义(P0.01),实验组少于对照组;应激后7 d两组仅中央活动时间差异有统计学意义(P0.05);(2)实验组小鼠进入开臂次数及停留时间在应激后立即、12 h较对照组均显著减少(P0.05);(3)实验组小鼠第2、3天的逃避潜伏期较对照组显著延长(P0.05),穿越平台次数及目标象限游程比例较对照组均显著增多(P0.05)。结论足底电击可引起小鼠自发行为减少,导致探索样行为减弱,焦虑增加,学习能力降低,但在一定程度上增强小鼠的空间记忆能力。
[Abstract]:Objective to study the effects of acute traumatic stress on spontaneous behavior, anxiety, learning and memory in mice.Methods Sixteen mice were divided into two groups: the experimental group and the control group. The experimental group was given electric shock on the plantar, the control group was given no treatment, the open field test and the elevated cross maze test were carried out at 12 hours and 7 days after treatment to evaluate the behavior of the mice.Water maze test was used to evaluate the changes of learning and memory in mice.Results (1) the number of times of standing and the time of central area activity in the two groups were significantly different between the two groups at 0 h and 12 h after stress, and the experimental group was less than the control group.7 days after stress, there was a significant difference in the time of central activity between the two groups (P 0.05) the number of times of entering the arm and the staying time of the mice in the experimental group immediately after stress decreased significantly compared with the control group for 12 h) the mice in the experimental group escaped from latent latency on the 2nd day of the third day.Compared with the control group, the time of crossing the platform and the proportion of the target quadrant run were significantly longer than that of the control group (P 0.05).Conclusion the plantar electric shock can reduce the spontaneous behavior, weaken the exploratory behavior, increase anxiety and decrease the learning ability, but to some extent enhance the spatial memory ability of mice.
