中药绞股蓝对血管性痴呆小鼠认知功能的保护作用

【摘要】:目的:探讨中药绞股蓝对脑缺血再灌注所致的血管性痴呆小鼠海马神经元损伤、认知功能障碍的保护作用及机制。 方法:采用双侧颈总动脉阻断(2VO)方法复制血管性痴呆小鼠模型,随机将雄性C57BL6/G小鼠分为5组,即假手术(sham)组、血管性痴呆模型(VD)组、绞股蓝治疗组(低、中、高剂量组)。行为学Morris水迷宫检测小鼠空间学习记忆能力、HE染色法观察小鼠海马神经元的形态学变化、酶联免疫分析法检测小鼠海马组织环磷酸腺苷(cAMP)的表达。 结果:①Morris水迷宫测试发现VD小鼠学习记忆能力下降;VD模型组小鼠的逃避潜伏期明显比Sham组时间长(P0.01),绞股蓝治疗组小鼠逃避潜伏期明显比VD模型组时间短(P0.05),绞股蓝中、高剂量组逃避潜伏期明显比低剂量组时间短(P0.05);绞股蓝中、高剂量组之间无显著差异(P0.05);VD模型组穿过平台次数明显少于Sham组(P0.01),VD模型组穿过平台次数明显少于GP低、中、高剂量组(P0.05),GP中、高剂量组穿过平台次数明显比低剂量组多(P0.05)。②HE染色显示,Sham组CA1区海马神经元细胞数目多,排列整齐紧密,细胞核呈圆形或椭圆形;VD模型组小鼠海马神经元胞浆出现片状坏死,海马神经细胞数目明显减少,排列紊乱松散,细胞核固缩深染,细胞核边界模糊。而GP中、高剂量组海马神经元细胞层次较清楚,排列较整齐,海马神经元细胞数量明显增多,细胞核固缩深染现象少见。③与假手术组比较,VD模型组小鼠海马组织cAMP含量明显降低(P0.01),与VD模型组比较,绞股蓝治疗低、中、高剂量组小鼠海马组织cAMP含量明显增高(P0.01),与绞股蓝治疗低剂量组比较,绞股蓝治疗中、高剂量组小鼠海马组织cAMP含量明显增高(P0.05)。 结论:VD小鼠海马区神经元损伤和脑认知功能减退;中药绞股蓝对VD小鼠的神经元损伤具有保护作用,增加海马组织cAMP的表达,有利于改善VD小鼠的学习记忆能力。
[Abstract]:Aim: to investigate the protective effect and mechanism of gynostemma pentaphyllum on hippocampal neuron injury and cognitive dysfunction in vascular dementia mice induced by cerebral ischemia and reperfusion.Methods: the vascular dementia model was established by bilateral common carotid artery occlusion in mice. Male C57BL6/G mice were randomly divided into 5 groups: sham group, vascular dementia model group, gynostemma gynostemma group (low, middle and high dose group).Behavioral Morris water maze was used to detect the spatial learning and memory ability of mice. The morphological changes of hippocampal neurons in mice were observed by HE staining and the expression of cyclic adenosine monophosphate (camp) was detected by enzyme-linked immunosorbent assay (Elisa).Results the escape latency of VD mice was significantly longer than that of Sham group (P 0.01), the escape latency of gynostemma treated group was shorter than that of VD model group (P 0.05), and that of gynostemma pentaphyllum group was lower than that of VD model group.The escape latency of high dose group was significantly shorter than that of low dose group (P 0.05), in gynostemma pentaphyllum, there was no significant difference between high dose group and high dose group. The number of passing through platform in high dose group was significantly less than that in Sham group (P 0.01) and the number of crossing platform in VD group was significantly less than that in GP.In the high dose group (P 0.05), the number of neurons passing through the plateau in the high dose group was significantly higher than that in the low dose group, and the number of hippocampal neurons in the CA1 area of the Sham group was more than that in the low dose group.In the group of round or elliptical VD model, the neurons in the hippocampus showed lamellar necrosis, the number of neurons in the hippocampus decreased obviously, the number of neurons in the hippocampus was loosely arranged, the nuclei were pyknotic and deep stained, and the nuclear boundaries were blurred.In GP group, the level of hippocampal neurons was clear and arranged neatly, and the number of hippocampal neurons increased obviously.Compared with sham-operated group, the cAMP content of hippocampus in VD model group was significantly lower than that in VD model group, and that in VD model group was lower than that in VD model group.The content of cAMP in hippocampus of high dose group was significantly higher than that of low dose group of gynostemma pentaphyllum. The content of cAMP in hippocampus of high dose group was significantly higher than that of low dose group of gynostemma pentaphyllum.Conclusion gynostemma pentaphyllum has a protective effect on neuron damage in VD mice, increases the expression of cAMP in hippocampus, and improves the learning and memory ability of VD mice.
