Caspase-3抑制剂干预甲基苯丙胺慢性依赖大鼠脑神经细胞凋亡与BCL-2表达的改变

【摘要】:目的观察z-DEVD-fmk对甲基苯丙胺(MAP)依赖大鼠行为变化的影响;脑组织细胞Caspase-3蛋白酶活性的变化;脑组织相关区域神经细胞凋亡及对BCL-2蛋白表达的影响。 方法建立甲基苯丙胺依赖大鼠实验动物模型,同时侧脑室注射z-DEVD-fmk进行干预,采用酶标仪法检测Caspase3酶活性;TUNEL法检测大鼠脑神经细胞凋亡;免疫组化技术检测BCL-2蛋白表达的影响。 结果 1.MAP慢性依赖大鼠100%出现异常行为表现,主要有异常不自主点头、竖毛自残、原地转圈;易激动等症状均有发生,给予z-DEVD-fmk干预后上述症状发生较少。 2. Caspase3酶活性:z-DEVD-fmk干预后与MAP慢性依赖组相比较Caspase-3酶活性降低,有显著性差异(P<0.05)。 3.大鼠脑组织细胞凋亡:z-DEVD-fmk干预下的MAP慢性依赖组大鼠脑组织细胞凋亡明显减少,与MAP慢性依赖组相比有显著性差异(P<0.01)。 4. BCL-2蛋白表达:免疫组织化学染色显示z-DEVD-fmk干预下,,MAP慢性依赖组大鼠脑组织细胞BCL-2阳性细胞数增加,与MAP慢性依赖组相比较有显著性差异(P<0.01)。 结论 1. z-DEVD-fmk干预后Caspase-3酶活性较MAP组降低; 2. z-DEVD-fmk干预后减少MAP慢性依赖大鼠脑组织细胞凋亡,上调BCL-2蛋白的表达。
[Abstract]:Objective to observe the effect of z-DEVD-fmk on the behavioral changes of rats with methamphetamine mitogen-dependent activity of Caspase-3 protease in brain tissue and the effect of neuronal apoptosis on the expression of BCL-2 protein in brain tissue related regions. Methods the experimental animal model of methamphetamine dependent rats was established, and z-DEVD-fmk was injected into the lateral ventricle to intervene. The activity of Caspase3 was detected by enzyme labeling method and Tunel method was used to detect the apoptosis of rat brain nerve cells, and the effect of BCL-2 protein expression was detected by immunohistochemical technique. Result Abnormal behavior was found in 100% of 1.MAP chronic dependent rats, including abnormal involuntary nodding, self-mutilation of hair, rotation in situ, agitation and other symptoms, and the occurrence of these symptoms was less after the intervention of z-DEVD-fmk. 2. The activity of Caspase3 enzyme: z-DEVD-fmk was significantly lower than that of chronic dependent MAP group (P < 0.05). 3. Apoptosis of brain tissue cells in MAP chronic dependent group was significantly decreased after intervention of z-DEVD-fmk (P < 0.01), which was significantly lower than that in MAP chronic dependence group (P < 0.01). 4. Expression of BCL-2 protein: immunohistochemical staining showed that the number of BCL-2 positive cells in brain tissue of rats with chronic dependence of map was increased after z-DEVD-fmk intervention, and there was a significant difference compared with that of chronic dependence group of MAP (P < 0.01). Conclusion 1. The activity of Caspase-3 in z-DEVD-fmk group was lower than that in MAP group. 2. After z-DEVD-fmk intervention, the apoptosis of brain cells and the expression of BCL-2 protein were decreased in chronic dependent MAP rats.
