首发抑郁症患者Notch通路及相关miRNAs的表达差异研究

【摘要】:目的:在人群水平探讨Notch通路相关的m RNAs、mi RNAs与抑郁症发生的关系。方法:采用病例对照研究设计,严格按照入排标准:1.实验一收集18名首发抑郁症患者和18名正常人群的外周血,提取RNA,通过实时荧光定量PCR(Quantitative real-time PCR,q RT-PCR)检测Notch信号通路中Notch1、RBP-jk、Numb、Hes1的m RNA表达水平,分析比较抑郁患者与正常人群外周血表达的差异、及其与认知功能等指标的相关性;2.实验二新收集32名首发抑郁症患者和32名正常人群,采集外周血提取RNA,通过q RT-PCR检测与Notch信号通路相关的mi RNAs(mi R-34b-5p、mi R-34c-5p、mi R-107、mi R-381、mi R-369-3p)以及m RNAs(Notch1、Hes1)的表达水平,分析比较抑郁症患者与正常人群外周血表达的差异,及其与临床症状、电生理、认知功能等指标的相关性。结果:实验部分一:1.抑郁组Notch1(Z=-3.354,P=0.001)表达低于对照组,RBP-jk(Z=-2.689,P=0.007)、Numb(Z=-3.101,P=0.002)表达高于对照组,未发现Hesl基因表达有差异(P0.05);2.抑郁症患不同亚型分组后,Notch1、RBP-jk、Numb、Hes1表达无差异;未发现Notch相关因子与认知功能存在相关性。实验部分二:1.抑郁组mi R-34b-5p(Z=-2.457,P=0.014)、mi R-34c-5p(Z=-2.001,P=0.045)表达高于正常组,抑郁组Notch1(Z=-2.021,P=0.043)的表达低于对照组;2.Notch1与mi R-34b-5p(r=-0.370,P=0.040)、mi R-34c-5p(r=-0.377,P=0.036)呈负相关;在mi RNAs之间,mi R-34b-5p与mi R-34c-5p(r=0.881,P0.001)呈高度正相关,mi R-34b-5p与mi R-107(r=0.794,P0.001)、mi R-381(r=0.877,P0.001)呈中、高度正相关,mi R-34c-5p与mi R-107(r=0.943,P0.001)、mi R-381(r=0.892,P0.001)呈高度正相关,mi R-107与mi R-381(r=0.835,P0.001)呈高度正相关。3.不同抑郁症亚型分组后,有自杀意念的抑郁症患者mi R-34b-5p(Z=-1.992,P=0.046)、mi R-369-3p(Z=-3.131,P=0.002)的表达都低于无自杀意念的患者。4.mi R-34b-5p(r=0.680,P0.001)、mi R-34c-5p(r=0.718,P0.001)、mi R-107(r=0.738,P0.001)、mi R-381(r=0.713,P0.001)与HAMD17评估得分呈正相关;mi R-34b-5p(r=0.481,P=0.015)、mi R-34c-5p(r=0.506,P=0.010)、mi R-107(r=0.503,P=0.010)、mi R-381(r=0.485,P=0.014)与N1潜伏期呈正相关,mi R-34b-5p(r=0.429,P=0.032)、mi R-34c-5p(r=0.479,P=0.016)、mi R-107(r=0.464,P=0.019)、mi R-381(r=0.566,P=0.003)与P2潜伏期呈正相关。结论:1.抑郁症患者存在外周血Notch通路相关m RNAs表达异常。2.抑郁症患者存在外周血Notch通路相关mi RNAs表达异常,且与抑郁症临床症状、认知功能密切相关。3.mi R-34b-5p、mi R-34c-5p可能靶向调控Notch参与抑郁症的发生。
[Abstract]:Objective: to investigate the relationship between Notch pathway related mRN Asmi RNAs and depression. Methods: a case-control study was designed, strictly according to the entry standard of 1: 1. In experiment 1, the peripheral blood of 18 patients with first-episode depression and 18 normal controls were collected, and the expression of m RNA in the Notch signaling pathway was detected by real-time fluorescence quantitative PCR(Quantitative real-time PCRQ RT-PCR1. To analyze and compare the difference of peripheral blood expression between depressive patients and normal subjects, and its correlation with cognitive function. In experiment 2, 32 first-episode depression patients and 32 normal controls were collected and the expression levels of mi RNAs(mi R-34b-5pttch1Hes1) and mRNAs-Notch1Hes1) related to Notch signaling pathway were detected by Q RT-PCR. To analyze and compare the difference of peripheral blood expression between depression patients and normal people, and its correlation with clinical symptoms, electrophysiology, cognitive function and so on. Results: the experiment part is 1: 1. The expression of Hesl gene in depression group was lower than that in control group. There was no significant difference in the expression of Numb-Hes1 in patients with different subtypes of depression, and no correlation between Notch related factors and cognitive function was found.
