AD患者血清中L-谷氨酰胺、溶血磷脂酰胆碱和棕榈酰胺HPLC-MS测定

【摘要】:正近年来寻找探索一种或几种特异、敏感的生物标志物,早期诊断阿尔茨海默病(Alzheimer disease,AD),及时干预和治疗,使其不发生或延缓发生受到国内外学者重视[1-2]。本课题组在采用超高效液相色谱-四级杆-飞行时间串联质谱仪研究筛选AD患者血清中生物标志物时发现L-谷氨酰胺(Lglutamine,LG)、溶血磷脂酰胆碱(lysophosphatidylcholine,LPC)和棕榈酰胺(palmitic amide,PA)与对照组有显著性差异。本文建立人血清中此标志物高效液相色谱质谱测定方法,为研究AD代谢通路和发病机制,早期诊断、预防和治疗提供科学依据。
[Abstract]:In recent years, we are looking for one or more specific and sensitive biomarkers, early diagnosis of Alzheimer disease AD, timely intervention and treatment, so that it does not occur or delay the occurrence of attention by domestic and foreign scholars [1-2]. In the study of screening serum biomarkers in AD patients by ultra-high performance liquid chromatography-quaternion-time-of-flight mass spectrometer, we found Lglutamine (LG), lysophosphatidylcholine (LPC) and palmitic amide-PA (palmitic amidePA) in serum of AD patients. There was significant difference between the two groups. A high performance liquid chromatography-mass spectrometry (HPLC / MS) method for the determination of this marker in human serum was established to provide scientific basis for the study of metabolic pathway and pathogenesis, early diagnosis, prevention and treatment of AD.
