利用基因芯片技术筛选汉族人群甲基苯丙胺依赖相关基因的初步研究

【摘要】:目的探索汉族群体中甲基苯丙胺依赖相关的差异表达基因。方法采集14例汉族甲基苯丙胺依赖者以及8例年龄、性别相匹配正常人的外周全血,利用mRNA表达谱芯片对其进行基因表达谱分析,并对差异表达基因进行功能分类分析。结果在27 958条转录本中,实验组和对照组差异表达倍数在2倍或2倍以上的转录本有3 560条,其中上调表达基因1 510个,下调表达基因2 576个。对这些差异基因分析发现甲基苯丙胺对细胞组分、分子功能、生物学过程的负调节有重要影响,并与转录调控、核苷酸代谢和修饰、蛋白质修饰、细胞凋亡、应激调节、自身免疫反应等相关。结论甲基苯丙胺成瘾的发生涉及多基因表达的改变,芯片技术可以有效地筛选出甲基苯丙胺依赖者与正常对照的差异表达基因,对进一步探索甲基苯丙胺成瘾机制、有效的干预或逆转毒品成瘾具有重要意义。
[Abstract]:Objective to explore the differentially expressed genes related to methamphetamine dependence in Han population. Methods Peripheral blood samples were collected from 14 patients with methamphetamine dependence in Han nationality and 8 healthy subjects of age and sex. The gene expression profiles were analyzed by mRNA microarray and the functional classification of differentially expressed genes was performed. Results among the 27 958 transcripts, there were 3 560 transcripts with 2 or more times of differential expression, including 1 510 up-regulated genes and 2 576 down-regulated genes. It was found that methamphetamine had an important effect on the negative regulation of cell composition, molecular function and biological process, and related to transcription regulation, nucleotide metabolism and modification, protein modification, cell apoptosis, stress regulation. Autoimmune response and so on. Conclusion the occurrence of methamphetamine addiction involves the change of polygene expression. Microarray technique can effectively screen differentially expressed genes between methamphetamine addicts and normal controls, and further explore the mechanism of methamphetamine addiction. Effective intervention or reversal of drug addiction is of great significance.
