不同程度慢性酒精中毒对大鼠内耳肽类激素的影响

【摘要】:目的:探讨不同程度的慢性酒精中毒对大鼠内耳肽类激素VIP(血管活性肠肽)及其受体VIPR、SP(P物质)及其受体(SPR)表达的影响及其机制。 方法:30只Sprague Dawley (SD)大鼠随机分为三组(轻度酒精中毒组、重度中毒组及对照组)进行造模。采用RT-PCR分别检测三组大鼠内耳VIP的表达,采用western blot检测大鼠内耳VIP受体蛋白的表达,采用免疫组化的方法定位及定量检测大鼠内耳VIP的表达。 结果:慢性酒精中毒组的大鼠内耳VIP及其受体的表达下调,并且重度慢性酒精中毒的大鼠内耳VIP及其受体的表达低于轻度组,差异有统计学意义(P0.05)。慢性酒精中毒组的大鼠内耳VIP及其受体与对照组在神经节细胞的表达差异无统计学意义(P0.05)。 结论: VIP及其受体、SP及其受体在慢性酒精中毒大鼠的听觉系统功能中发挥着重要的作用。慢性酒精中毒可引起听觉系统的一种肽类激素分泌不平衡,,最终导致听力损失。
[Abstract]:Aim: to investigate the effect and mechanism of chronic alcoholism on the expression of vasoactive intestinal peptide (VIP) and its receptor VIPR,SP (substance P) and its receptor (SPR) in rat inner ear. Methods: thirty Sprague Dawley (SD) rats were randomly divided into three groups: mild alcoholism group, severe intoxication group and control group. The expression of VIP in the inner ear of the three groups was detected by RT-PCR, the expression of VIP receptor protein in the inner ear of the rat was detected by western blot, and the expression of VIP in the inner ear of the rat was detected quantitatively by immunohistochemical method. Results: the expression of VIP and its receptor in the inner ear of the rats with chronic alcoholism was down-regulated, and the expression of VIP and its receptor in the inner ear of the rats with severe chronic alcoholism was lower than that of the mild group (P0.05). There was no significant difference in the expression of VIP and its receptor between the chronic alcoholism group and the control group in ganglion cells (P0.05). Conclusion: VIP and its receptors, SP and their receptors play an important role in the auditory system function of chronic alcoholism rats. Chronic alcoholism can cause an imbalance in the secretion of peptide hormones in the auditory system, leading to hearing loss.
