Alzheimer病模型小鼠海马代谢物早期变化的9.4T磁共振频谱研究

目的:通过9.4T磁共振频谱离体检测阿尔茨海默病(Alzheimer’s disease,AD)模型小鼠疾病过程中的代谢物变化,以找出MRS早期诊断此病的生物学标识。方法: 36只昆明小鼠随机分成5个实验组和1个对照组,每组6只,对于实验组各组小鼠每天进行皮下注射D-半乳糖120mg/(kg.d)和亚硝酸钠90mg/(kg.d),各组分别连续注射30天、40天、50天、60天及70天,对照组小鼠每天注射相同剂量的生理盐水70天,药物注射结束后行水迷宫测试(MWM)并牺牲小鼠取其两侧海马。右侧海马经高氯酸萃取等处理后冻干成粉末,利用9.4T磁共振进行频谱测试观察代谢物变化;左侧海马用来做病理组织学检测。对于代谢物氮-乙酰天门冬氨酸(N-acetylaspartate, NAA),谷氨酸(glutamate, Glu),肌醇(myo-inositol, mI)和胆碱(choine, Cho)进行分析。结果:水迷宫检查结果显示实验组70天时空间学习及记忆能力较对照组明显降低(P<0.05)。电镜下观察病理染色图片可见60天及70天之小鼠海马区神经元细胞明显减少,变性,神经原纤维缠结及老...
