丰富环境对妊娠期暴露脂多糖加速母鼠衰老相关认知功能及海马突触可塑性相关蛋白改变的影响

背景:衰老相关认知功能损害的机制及有效的治疗方法仍亟待解决。认知功能损害受许多不良因素如应激、感染、不良妊娠的影响,而丰富环境(EE)可能改善动物健康状况。证据表明突触可塑性是中枢神经系统学习和记忆的基本神经机制,对认知功能尤为重要,而突触可塑性相关蛋白的改变会导致突触可塑性改变从而影响认知功能。我们既往研究表明不良的妊娠环境(LPS暴露)会加剧母鼠老年期学习记忆能力的损害,但不知EE处理可否减轻这种改变以及一些重要的突触可塑性相关蛋白如突触后致密物95(PSD-95)、谷氨酸AMPA受体GluA1亚基(GluA1)和Homer1b/c蛋白是否涉及这些改变。目的:探讨妊娠期暴露LPS对CD-1母鼠衰老相关性认知功能的影响及EE对其的改善作用;评估海马区PSD-95、GluA1和Homer1b/c蛋白表达的衰老相关性改变及LPS和EE的效应;探讨认知功能与海马突触可塑性相关蛋白间的相关性。方法:CD-1母鼠在妊娠15-17天时随机连续每天腹腔注射LPS(50μg/kg)或等容积生理盐水(对照组)。母鼠正常分娩并在断奶后与子鼠分开。接受LPS的母鼠再分别接受EE饲养(LPS-E组)或普通饲...
