深圳市军团菌污染现况及分子流行病学研究

【摘要】:70年代美国爆发“军团病”以来,已经有多个国家有“军团病”的报道。我国南京在1982年首次证实军团菌病病例。多个城市对空调冷却塔进行军团菌调查,发现其中军团菌的阳性率为50%-80%。2003年中国经历SARS的袭扰后,人们对空调系统的卫生状况表现出空前的关注。本课题拟对深圳市空调冷却塔微生物污染状况,尤其是军团菌污染状况作较为全面的调查,建立流行病学资料。 目的:1.对深圳市宾馆、医院、影院、写字楼等公共场所的空调冷却塔水进行军团菌的分离培养,了解军团菌的污染状况。检测细菌总数、大肠菌群、粪大肠菌群,初步了解空调冷却塔微生物污染状况以及军团菌与它们之间是否存在一定联系。2.对所采集的冷却塔水进行PCR检测,并与常规分离培养方法进行比较。3.用AFLP技术对常规分离培养出的菌株进行分子生物学分型,分析分离菌株的多态性,确定菌株的遗传基础之间相互的联系和变异,了解深圳市军团菌分布流行病学特征。 方法:1.采用平皿计数法对中央空调冷却塔水进行军团菌的分离培养,用血清学方法对分离菌株进行初步鉴定。2.检测冷却塔水中
[Abstract]:Since the outbreak of Legionnaires disease in the United States in the 1970s, there have been reports of Legionnaires' Disease in many countries. Legionella was first confirmed in 1982 in Nanjing, China. Legionella in air conditioning cooling towers was investigated in many cities. It was found that the positive rate of Legionella was 50%-80%. After China experienced the disturbance of SARS in 2003, people showed unprecedented concern about the sanitary condition of air conditioning system. The aim of this paper is to investigate the microorganism pollution of air conditioning cooling tower in Shenzhen, especially Legionella, and to establish epidemiological data. Purpose 1. The air conditioning cooling tower water of Shenzhen hotels, hospitals, cinemas, office buildings and other public places were isolated and cultured to understand the contamination of Legionella. The total number of bacteria, coliform group, fecal coliform group were detected to understand the microorganism pollution in cooling tower of air conditioning system and whether there was a certain relationship between Legionella and them. The collected cooling tower water was detected by PCR and compared with conventional culture method. 3. 3. The molecular biological typing of the strains isolated by conventional methods was carried out by using AFLP technique, the polymorphism of the isolates was analyzed, the relationship and variation of the genetic basis of the strains were determined, and the epidemiological characteristics of the distribution of Legionella in Shenzhen were understood.
