一种固体二氧化氯杀灭微生物效果的研究

【摘要】:二氧化氯(简写为ClO_2)被公认为一种广谱、高效、快速、低毒的消毒剂,已广泛应用于环境物体表面、食品加工设备、餐饮容器与水的消毒等各方面。近年来,重庆市申报ClO_2消毒剂的生产厂家逐年增加,剂型种类繁多,产品标准和质量参差不齐。我们在实际工作中发现,单纯用国家检测规范中的杀菌效果指标进行同类产品的鉴定不能完全体现ClO_2消毒剂的特点,不能明确描述其质量高低,不利于指导和监督厂家提高产品质量,给消毒剂使用单位推荐产品时也缺乏科学的依据。因此,在实验室对康保洁牌二氧化氯固体消毒剂(简写为康保洁ClO_2)进行了杀菌效果的系统分析,观察其特性和杀菌效果变化规律,从多方面评价该消毒剂的质量,探讨同类产品可能增加的有效评价指标。也借此工作为ClO_2消毒剂质量的系统评价奠定实验基础。 为了解康保洁ClO_2的杀菌性能、稳定性及其影响因素,我们在实验室对其进行了不同活化条件下的ClO_2含量测定、悬液定量杀灭试验、杀菌影响因素测定、加速稳定性试验、能量试验,并通过现场与模拟现场消毒试验验证了该消毒剂的实用消毒效果。采用气相色谱法测定其消毒副产物,验证ClO_2消毒的优越性。研究结果如下: 1. 康保洁ClO2 为二元包装,A 剂主要成分为亚氯酸钠与稳定剂碳酸钠,呈白色粉末状,其中亚氯酸钠含量≥82%,B 剂为活化剂柠檬酸,为白色细小颗粒。使用时取等量A 剂、B 剂加入适量水活化一定时间,亚氯酸盐在柠檬酸的激活下释放出具有活性功能的游离ClO_2。ClO_2释出浓度随活化水量增多而降低,活化时间的延长而增高,活化至4h 后下降。经杀菌试验验证, 1.0gA 剂与1.0gB 剂以5.0ml 水活化4h,此时能释出ClO2 90.79mg/g,与不同活化条件的康保洁ClO_2相比较,其杀灭微生物作用最强。 2. 康保洁ClO_2 属稳定类消毒剂,具有高效、快速的杀菌特点。225mg/L 康保洁ClO2 对细菌芽胞作用2min 或150mg/L 作用15min,5mg/L 对铜绿假单胞菌作用1min,7.5mg/L 对金黄色葡萄球菌作用1min 或5mg/L 作用3min,7.5mg/L 对大肠杆菌作用3min,杀灭对数值均≥5.00,7.5mg/L 康保洁ClO_2 对白色念珠菌作用3min 或5mg/L作用7min,其杀灭对数值均≥4.00,达消毒合格要求。 3. 康保洁ClO_2 可用于多次消毒污秽物品,其实用参考剂量为10mg/L。5mg/L 康
[Abstract]:Chlorine dioxide (ClO_2) has been recognized as a broad-spectrum, efficient, rapid, low-toxic disinfectant, has been widely used in the surface of environmental objects, food processing equipment, catering containers and water disinfection and so on. In recent years, the manufacturers of ClO_2 disinfectant declared by Chongqing have increased year by year, the dosage forms are various, and the product standard and quality are not uniform. In our practical work, we found that the identification of the same kind of products with the germicidal efficacy index in the national standard can not fully reflect the characteristics of the ClO_2 disinfectant, and can not clearly describe the quality of the disinfectant. It is not conducive to guide and supervise manufacturers to improve the quality of products, and there is no scientific basis for recommending products to disinfectant users. Therefore, the germicidal efficacy of Kangbaojie brand chlorine dioxide solid disinfectant (ClO_2) was systematically analyzed in the laboratory, and its characteristics and bactericidal effect were observed, and the quality of the disinfectant was evaluated from many aspects. This paper discusses the effective evaluation index which may be increased by the same kind of products. This work also lays an experimental foundation for the systematic evaluation of the quality of ClO_2 disinfectant. In order to understand the germicidal performance, stability and influencing factors of Kangpaojie ClO_2, we measured the content of ClO_2 under different activation conditions, the suspension quantitative killing test and the influence factors of germicidal activity in the laboratory. Accelerated stability test, energy test, and field and simulated disinfection tests verified the practical disinfection effect of the disinfectant. Gas chromatography was used to determine its disinfection by-products to verify the advantages of ClO_2 disinfection. The results of the study are as follows: 1. Kangpaojie ClO2 is a binary package. The main composition of agent A is sodium chlorite and stabilizer sodium carbonate, in which the content of sodium chlorite is more than 82%, and agent B is citric acid, which is a fine white particle. The release concentration of free ClO_2.ClO_2 with active function decreased with the increase of activated water amount, and the release concentration of free chlorite with active function was decreased with the increase of activated water amount, and the release concentration of free chlorite with active function was decreased with the increase of activated water quantity. The activation time increased and decreased after 4 h activation. The germicidal test showed that 1.0gA and 1.0gB were activated with 5.0ml water for 4 h, and then ClO2 90.79 mg 路kg-1 was released. Compared with Kangpaojie ClO_2 with different activation conditions, its bactericidal effect was the strongest. 2. Kangpaojie ClO_2 is a stable disinfectant with high efficiency and rapid bactericidal properties. 225 mg/L Kangpaojie ClO2 treated bacterial spores with 2min or 150mg/L for 15 min and 5 mg / L against Pseudomonas aeruginosa for 1 min.
