四君子汤对慢性心衰大鼠心肌组织蛋白质组学影响的研究

【摘要】:目的:探讨四君子汤对慢性心衰大鼠心肌组织蛋白质表达变化的影响。方法:SD雄性大鼠42只,随机取6只作为正常组,其余36只复制慢性心力衰竭大鼠模型,造模成功后,分为四君子汤高、低剂量组(11.88,2.97 g·kg~(-1)·d~(-1)),卡托普利组(100 mg·kg~(-1)·d~(-1)),正常组和模型组分别给予相同体积生理盐水,连续灌胃给药1月。分别留取各组左心室组织,双向凝胶电泳分离心肌蛋白,分析差异显示的蛋白,并对有显著差异的蛋白点进行胶内酶切和质谱鉴定。结果:与模型组比较,四君子汤组有31种蛋白质表达上调,28种蛋白质表达下调,各选取8个差异显著(Volume值相差≥1.5倍)的蛋白进行质谱鉴定,通过整合去重,鉴定出4个蛋白:与能量代谢相关的Suclg2蛋白、丙酮酸脱氢酶复合体;与心肌细胞收缩功能相关的肌球蛋白轻链2;与心肌细胞重构相关的丝氨酸蛋白酶抑制剂A3N。结论:四君子汤可调节慢性心衰大鼠心肌组织多种蛋白表达,可能通过调节能量代谢、减少心肌重构、提高收缩功能而改善心功能。
[Abstract]:Objective: to investigate the effect of Sijunzi decoction on the changes of myocardial protein expression in chronic heart failure rats. Methods: 42 male SD rats were randomly selected as normal group and the other 36 rats were duplicated with chronic heart failure model. The rats were divided into four groups: Sijunzi decoction was high, low dose group was 11.88g 路kg ~ (-1)) 路D ~ (-1), captopril group was 100 mg 路kg ~ (-1) 路d ~ (-1) ~ (-1), normal group and model group were given the same volume of normal saline respectively on January, left ventricular tissue was isolated by two-dimensional gel electrophoresis, myocardial protein was isolated by two-dimensional gel electrophoresis.
