养心汤对慢性心衰模型大鼠血清ET-1水平的影响

【摘要】：目的：本课题通过建立慢性心衰大鼠模型,观察养心汤对慢性心衰大鼠血清内皮素-1(ET-1)的水平和左室短轴缩短率(LVFS)的影响,以及心肌组织病理学的改变,探讨养心汤治疗慢性心衰大鼠的作用机制。方法：将60只SD大鼠随机选取10只作为空白对照组,其余50只用阿霉素连续腹腔注射进行造模,6周后将造模成功的大鼠随机平均分为3组,每组14只,进行灌胃给药。养心汤组给予养心汤悬浊液剂量2.34g/kg/d灌胃；卡托普利组给予卡托普利悬浊液剂量2.25mg/kg/d灌胃；空白对照组与模型对照组给予蒸馏水2ml/d灌胃。4周后,给予大鼠腹腔注射水合氯醛(3.5ml/kg)麻醉,使用超声诊断仪进行心功能测定后,取血5m1用以检测血清内皮素1的浓度,取大鼠心肌组织染色进行病理观察。结果：1.与模型对照组比较,养心汤组和卡托普利组大鼠的血清ET-1浓度水平有所下降,差异有统计学意义(P0.05)；养心汤组与卡托普利组相比,差异无统计学意义(P0.05)。2.与模型对照组比较,养心汤组和卡托普利组大鼠的LVFS有所增加,差异有统计学意义(P0.05)；养心汤组与卡托普利组比较,差异无统计学意义(P0.05)。3.与模型对照组比较,养心汤组和卡托普利组均能减轻心衰大鼠心肌组织的病理损伤。结论：1.养心汤能够降低慢性心衰大鼠血清ET-1的水平,其作用与卡托普利相当。2.养心汤能够提高心衰大鼠的LVFS,其作用与卡托普利相当。3.养心汤对慢性心衰大鼠心肌损伤组织有一定的修复作用,并且与卡托普利相当。
[Abstract]:Objective: to observe the effect of Yangxin decoction on serum endothelin-1 (ET-1) and left ventricular shortening rate (LVFSs) in rats with chronic heart failure (CHF), and to observe the pathological changes of myocardium in rats with chronic heart failure (CHF). To explore the mechanism of Yangxin decoction in the treatment of chronic heart failure rats. Methods: 60 SD rats were randomly selected as the blank control group. The other 50 rats were randomly divided into 3 groups with 14 rats in each group after 6 weeks of continuous intraperitoneal injection of adriamycin. The captopril group was given a dose of 2.25 mg / kg / d of captopril suspension, the blank control group and the model control group were given 2 ml / d distilled water for 4 weeks, then the rats were anesthetized by intraperitoneal injection of chloral hydrate (3.5 ml / kg), and the cardiac function was measured by ultrasonic diagnostic apparatus. The concentration of serum endothelin 1 was detected by blood 5m1, and the myocardial tissue staining of rats was taken for pathological observation. Results compared with the model control group, the serum ET-1 level in the Yangxin decoction group and captopril group was lower than that in the model control group. There was no significant difference between Yangxin decoction group and captopril group. Compared with model control group, the LVFS of Yangxin decoction group and captopril group was increased. There was no significant difference between Yangxin decoction group and captopril group, and there was no significant difference between Yangxin decoction group and captopril group. There was no significant difference between Yangxin decoction group and captopril group, and there was no significant difference between Yangxin decoction group and captopril group. Both Yangxin decoction group and captopril group can reduce the pathological damage of myocardial tissue in rats with heart failure. Conclusion: 1. Yangxin decoction can reduce the level of serum ET-1 in chronic heart failure rats. The effect of Yangxin decoction was similar to that of captopril. 2. Yangxin decoction could improve LVFSof heart failure rats, and its effect was similar to that of captopril. Yangxin decoction had certain repair effect on myocardial injury tissue of chronic heart failure rats, and was similar to captopril.
