虎潜丸对去卵巢大鼠骨密度及BMP-7水平影响的研究

【摘要】:目的:通过检测虎潜丸干预对去卵巢大鼠骨密度及骨组织中BMP-7水平的影响,探讨虎潜丸防治骨质疏松症的疗效。 方法:将6个月龄的40只雌性SD大鼠随机分为手术模型组、雌激素组、虎潜丸组和假手术组,每组10只。采用10%水合氯醛溶液对SD大鼠按0.33mL/100g体质腹腔注射进行麻醉,麻醉生效后,在无菌条件下,前三组由背侧入路,肋弓下缘备皮,络合碘消毒,切开皮肤,在脊柱旁开0.5cm与肋弓下缘0.5cm的交点处,进入腹腔剪去卵巢。另侧同法切除。假手术组大鼠也行同样手术入路打开腹腔,只切除腹膜后少许脂肪组织,不切除卵巢。切口内滴入少量青霉素水溶液,分层缝合手术切口。术后分笼饲养,用5万U单位青霉素注射液腹腔注射3d抗感染。术后4周,从造模组及假手术组中随机各选出6只大鼠行活体骨密度检查,以确定造模成功。在确定造模成功后,虎潜丸组给予虎潜丸煎剂浓缩液灌胃,每天1次;雌激素组给予尼尔雌醇悬浊液灌胃,每周1次;手术模型组和假手术组给予蒸馏水灌胃,每天1次。灌胃治疗12周后测量各组大鼠的骨密度及骨组织中BMP-7含量。 结果:手术模型组大鼠的骨密度低于假手术组,其差异具有统计学意义(P0.05),说明骨质疏松大鼠模型已建立;虎潜丸组及雌激素组大鼠骨密度均高于手术模型组,其差异具有统计学意义(P0.05),说明两种治疗方式均有治疗效果;虎潜丸组与雌激素组大鼠骨密度相比较无明显差距,无统计学意义(P0.05)。BMP-7阳性表达方面,虎潜丸组及雌激素组大鼠骨组织中BMP-7水平均高于手术模型组,其差异具有统计学意义(P0.05),虎潜丸组与雌激素组大鼠BMP-7水平相比较无明显差距,无统计学意义(P0.05) 结论:虎潜丸对切除卵巢后所致的骨质疏松症有明显的治疗作用,其机制可能是在一定程度地提高了组织中BMP-7的水平,从而抑制骨组织的吸收,促进骨组织的形成。
[Abstract]:Objective: to investigate the effect of Huqian pill on bone mineral density (BMD) and BMP-7 level in bone tissue of ovariectomized rats. Methods: forty female Sprague-Dawley rats aged 6 months were randomly divided into three groups: estrogen group, Huqian pill group and sham-operation group, 10 rats in each group. 10% chloral hydrate solution was used to anesthetize SD rats by intraperitoneal injection of 0.33 mL / 100 g constitution. After the anaesthesia took effect, under sterile conditions, the first three groups were treated by dorsal approach, the lower margin of the costal arch was covered with skin, the skin was disinfected with iodine, the skin was cut open, and the intersection between 0.5 cm of the spine and 0.5 cm of the lower margin of the rib arch was opened at the side of the spine. The rats in the sham-operation group also underwent the same operation to open the abdominal cavity, only a little fat tissue was removed from the peritoneum, and the ovary was not removed. A small amount of penicillin solution was dripped into the incision. After operation, 50,000 U penicillin injection was injected intraperitoneally for 3 days to resist infection. At 4 weeks after operation, 6 rats were randomly selected from the model group and sham-operation group to perform bone mineral density examination in vivo. In order to confirm the success of model making, the group of Huqian pills was given concentrated decoction of Huqian pills once a day, the estrogen group was given Neilestriol suspension once a week. Rats in the operation model group and the sham operation group were given distilled water once a day. The bone mineral density (BMD) and the content of BMP-7 in bone tissue were measured after 12 weeks of gastric perfusion. Results: the BMD of the rats in the operation model group was lower than that in the sham operation group, and the difference was statistically significant (P 0.05), which indicated that the osteoporosis rat model had been established, the bone mineral density of the rats in the Huqian pill group and the estrogen group were higher than that in the operation model group. There was no significant difference in bone mineral density (BMD) between the two groups, and there was no significant difference in BMD between the two groups, and there was no significant difference in the positive expression of P0.05U. BMP-7 between the two groups, and there was no significant difference in bone mineral density between the two groups, and there was no significant difference in the expression of BMD between the two groups. The level of BMP-7 in the bone tissue of rats in the Huqian pill group and estrogen group was higher than that in the operation model group, and the difference was statistically significant (P 0.05). There was no significant difference in the level of BMP-7 between the Huqian pill group and the estrogen group (P 0.05). Conclusion: Huqian pill has obvious therapeutic effect on osteoporosis caused by ovariectomy, and its mechanism may be to increase the level of BMP-7 in tissues to a certain extent, thus inhibit the absorption of bone tissue and promote the formation of bone tissue.
