前列灵胶囊对慢性前列腺炎炎症调控网络及氧化应激水平的影响实验研究

【摘要】:[目的]:通过前列灵胶囊对慢性前列腺炎大鼠治疗作用的实验研究,评价其治疗效果,探讨前列灵胶囊对慢性前列腺炎炎症调控网络及氧化应激水平的影响,为治疗慢性前列腺炎中药的开发应用奠定实验和理论基础。 [方法]:将60只大鼠驯化喂养一周后,随机分为6组,即空白组、阳性对照组、模型组、前列灵胶囊高剂量组、中剂量组、低剂量组。每组10只,各组用25%乌拉坦1gmg量,经腹腔注射麻醉,无菌条件下取下腹正中切口,打开腹腔,提出膀胱及两侧精囊,暴露前列腺背叶。其中空白组分别经前列腺双侧背叶注入无菌注射用水O.1ml,其余各组分别注入25%消痔灵注射液0.1ml,缝合后恢复1周。 给药方法:空白组和模型组每日灌胃一次,每次灌胃与实验组等剂量的生理盐水;阳性对照组(前列通瘀胶囊):每只大鼠经灌胃给予前列通瘀胶囊混悬液剂量相当于60kg成人用量20倍;前列灵高、中、低剂量组:每组大鼠给予前列灵胶囊混悬液灌胃,剂量分别相当于60kg成人用量20倍、10倍、5倍;均每日灌胃一次;连续给药30天,每天观察并记录大鼠活动、体重及精神状态变化,于末次给药后24小时,称每只大鼠的体重,每只大鼠采股动脉血3-5m1,离心冷藏,断头处死,摘除前列腺组织,前列腺组织用4%甲醛固定液固定。采用ELISA法分析各组大鼠血清中IL-6、ACP和SOD、MDA的含量。用S-P法检测前列腺组织中GSH、GST、GSH-px含量,用电镜观察前列腺组织中亡因子Bax.Bcl-2的变化。 [结果]:前列灵胶囊可降低前列腺炎大鼠血清中IL-6.ACP.MDA和升高SOD的含量,以及升高慢性前列腺炎大鼠前列腺组织中GSH. GST.GSH-px的含量,前列灵胶囊还具有上调Bax蛋白含量和降低Bcl-2蛋白含量的作用。 [结论]:前列灵胶囊对大鼠慢性前列腺炎有明显的治疗作用,其作用机理与降低前列腺炎大鼠血清中IL-6、ACP、MDA和升高SOD含量有关,以及升高慢性前列腺炎大鼠前列腺组织中GSH.GST. GSH-px的含量有关,前列灵胶囊还具有上调Bax蛋白含量和降低Bcl-2蛋白含量的作用。
[Abstract]:[objective]: to evaluate the therapeutic effect of Qianlieling capsule on chronic prostatitis rats, and to explore the effect of Qianlieling capsule on inflammatory regulatory network and oxidative stress level in chronic prostatitis rats. To lay an experimental and theoretical foundation for the development and application of traditional Chinese medicine for the treatment of chronic prostatitis. [methods] Sixty rats were randomly divided into 6 groups: blank group, positive control group, model group, high dose group, middle dose group, low dose group, 10 rats in each group, and each group was given 25% uratan 1 g mg, and 10 rats in each group were randomly divided into 6 groups, namely the blank group, the positive control group, the model group, the high dose group of Qianlieling capsule, the middle dose group and the low dose group. Anesthetized by intraperitoneal injection, the median incision of the lower abdomen was taken under aseptic conditions, the abdominal cavity was opened, and the bladder and seminal vesicles were put forward. In the blank group, 0.1 ml of water was injected into the bilateral dorsal lobe of the prostate, and 0.1 ml of 25% Xiaozhiling injection was injected into the rest groups, and then recovered for 1 week after suture. Methods: the blank group and the model group were given intragastric once a day, and each time they were given the same dose of normal saline as the experimental group. The positive control group (Qianlie Tongyu capsule: the dosage of Qianlie Tongyu capsule suspension was 20 times that of 60 kg adult rats, the dosage of Qianlie Ling high, middle dose and low dose group: each group rats were given Qianliesling capsule suspension, the dosage of Qianlie Tongyu capsule suspension was 20 times as much as 60 kg adults, the rats of each group were given Qianlieling capsule suspension, The doses were 20 times as much as 10 times as much as 5 times as much as 60 kg of adults. They were given orally once a day. For 30 consecutive days, the rats' activities, body weight and mental state were observed and recorded every day, and the weight of each rat was weighed 24 hours after the last administration. The femoral arterial blood was collected from each rat from 3 to 5 m1.The rats were cryopreserved centrifugally, killed by decapitation, and the prostate tissue was removed. The contents of IL-6ACP and SDD in serum of rats in each group were analyzed by ELISA method. The GSH-px content in prostate tissue was detected by S-P method. The changes of Bax.Bcl-2 in prostate tissue were observed by electron microscope. [results]: Qianlieling capsule could decrease the content of IL-6.ACP.MDA and increase the content of SOD in serum of rats with prostatitis, and increase the content of GSH. GST.GSH-px in prostate tissue of rats with chronic prostatitis. Qianlieling capsule can also up-regulate the content of Bax protein and decrease the content of Bcl-2 protein. [conclusion]: Qianlieling capsule has obvious therapeutic effect on chronic prostatitis in rats, and its mechanism is related to the decrease of IL-6 ACP MDA and the increase of SOD content in serum of rats with prostatitis. The content of GSH.GST. GSH-px in prostatic tissue of rats with chronic prostatitis was increased, and the content of Bax protein was up-regulated and the content of Bcl-2 protein was decreased by Qianlieling capsule.
