白豆蔻挥发油对糖尿病肾病大鼠肾脏保护作用

【摘要】:目的:研究白豆蔻挥发油对糖尿病肾病大鼠肾脏保护作用及其机制。方法:46只Wistar大鼠随机分为正常组(6只)和模型组(40只),模型组以腹腔注射STZ建立糖尿病肾病模型,造模后随机分模型组,白豆蔻低、高剂量组,六味地黄丸组4组,各10只。连续灌胃给药21d,每周检测1次血糖;最后一次给药24h后麻醉大鼠腹主动脉取血,检测血清肌酐(Cre)、尿素氮(BUN)、白蛋白(ALB)含量;光镜下观察肾脏组织病理改变;免疫组化法检测基质金属蛋白酶2(MMP-2)、转化生长因子β1(TGF-β1)、胰岛素样生长因子2(IGF-2)的表达。结果:与正常组比较,模型组血糖显著升高(P0.01),经过给药后各给药组血糖不同程度下降,其中白豆蔻高剂量组血糖显著下降(P0.05,P0.01);与模型组比较,各给药组Cre、BUN、ALB含量均降低,其中白豆蔻高剂量组作用明显(P0.05,P0.01)。光镜下病理形态学观察显示,各给药组对大鼠肾组织损伤有不同程度的改善作用;各给药组MMP-2、TGF-β1、IGF-2蛋白表达均上调。结论:白豆蔻挥发油对肾脏有一定的保护作用,作用机制可能与上调MMP-2、TGF-β1、IGF-2的表达有关,从而改善链脲佐菌素所致糖尿病肾病的病理改变。
[Abstract]:Objective: to study the protective effect and mechanism of essential oil of cardamom cardamom on the kidney of diabetic nephropathy rats. Methods: 46 Wistar rats were randomly divided into normal group (n = 6) and model group (n = 40). Diabetic nephropathy model was established by intraperitoneal injection of STZ in the model group. After the model was established, the rats were randomly divided into model group, low dose group, high dose group and Liuwei Dihuang pill group, 10 rats in each group. Blood glucose was measured once a week for 21 days, and blood was taken from abdominal aorta of anesthetized rats 24 hours after the last administration. Serum creatinine (creatinine), urea nitrogen bun (bun), albumin (ALBB) were detected; pathological changes of renal tissue were observed under light microscope; expression of matrix metalloproteinase 2 (MMP-2), transforming growth factor 尾 1 (TGF- 尾 1), insulin-like growth factor 2 (IGF-2) was detected by immunohistochemical method. Results: compared with normal group, The blood glucose of the model group increased significantly, and the blood glucose of each drug group decreased in varying degrees after administration, among which, the blood glucose of the high dose group decreased significantly (P0.05P0.01A), and compared with the model group, the ALB content of creatine BUNA in each administration group decreased, and the blood glucose of each group decreased in different degrees, compared with the model group, the blood glucose in the high dose group decreased significantly. Among them, the high dose group of cardamom had obvious effect on P0.05 and P0.01.The histopathologic observation under light microscope showed that each group had different degree of ameliorative effect on the renal tissue injury of rats. Conclusion: the essential oil of cardamom has a protective effect on the kidney, and the mechanism may be related to the up-regulation of the expression of MMP-2TGF- 尾 1IGF-2, thus improving the pathological changes of streptozotocin induced diabetic nephropathy.
