HAT抑制剂Garcinol对内毒素诱导的急性肝损伤的保护作用及机制研究

【摘要】:目的:乙酰化修饰是蛋白质翻译后修饰(post-translationalmodification, PTM)的重要方式之一,可发生在组蛋白和多种炎症相关转录因子,是调控炎症相关基因表达的重要机制。乙酰化修饰是可逆的,乙酰化和去乙酰化修饰分别由组蛋白乙酰转移酶(histoneacetyltransferases, HATs)和组蛋白去乙酰化酶(histone deacetylases,HDACs)所催化。目前关于HDACs抑制剂对炎症的调节效应已有较多研究,但HATs抑制剂是否也可影响炎症的发生发展却少有报道。Garcinol是常用的HATs抑制剂,本文在脂多糖(lipopolysaccharide,LPS)/右旋半乳糖胺(D-galactosamine, D-Gal)诱导的小鼠急性肝损伤模型中观察了garcinol对肝损伤的调节效应并初步探讨了其可能的机制。 方法:雄性BALB/c小鼠腹腔注射LPS(10μg/kg)及D-Gal(700mg/kg)诱导急性爆发性肝损伤,溶剂或不同剂量garcinol(2.5mg/kg,5mg/kg,10mg/kg)在LPS/D-Gal处理前0.5h经腹腔注入。LPS/D-Gal处理后1.5h,小鼠经麻醉后断颈处死,采集血浆及肝组织样本,ELISA法检测各样本中TNF-α和IL-6的水平。另一批小鼠在LPS/D-Gal处理后6h处死并采集血浆及肝组织样本。采用化学显色法检测血浆中转氨酶ALT和AST水平、测肝组织中丙二醛(malondialdehyde, MDA)含量及caspase-3,-8,-9活性;免疫印迹法检测肝组织中p65,乙酰化p65,p53,乙酰化p53,激活型caspase-3,,Bax及Bcl-2蛋白含量;肝组织经福尔马林固定后,常规HE染色观察组织病理学改变,TUNEL法检测细胞凋亡,免疫组织化学法检测激活型caspase-3水平。 结果:研究发现HATs抑制剂garcinol可明显减轻LPS/D-Gal诱导的肝损伤,garcinol对模型小鼠肝内炎症反应无明显影响但可显著抑制肝细胞凋亡。具体表现在:(1)Garcinol在2.5~10mg/kg范围内能剂量依赖性抑制LPS/D-Gal诱导的血浆中ALT和AST的升高,这一效应伴随有模型小鼠肝组织内MDA水平的降低及组织病理损伤的减轻;(2)LPS/D-Gal可诱导肝组织内NF-κB p65亚单位上Lys310位点的乙酰化修饰,这可被garcinol所抑制,但garcinol对LPS/D-Gal诱导的促炎性细胞因子TNF-α和IL-6的表达并无明显影响;(3)TUNEL分析发现garcinol可显著抑制LPS/D-Gal诱导的肝细胞凋亡,这伴随有肝组织内激活型caspase-3水平的降低及caspase-3,-8,-9活性的下调;(4)Garcinol也可明显抑制LPS/D-Gal诱导的肝组织内p53上Lys379位点的乙酰化修饰、下调促凋亡蛋白Bax的表达,但garcinol对抗凋亡蛋白Bcl-2的含量无明显影响。 结论:本研究数据显示HATs抑制剂garcinol对LPS/D-Gal诱导的肝损伤具有保护作用,这一保护效应不伴随有肝内炎症因子的表达降低,可能通过抑制p53的乙酰化修饰、降低其活性,从而下调促凋亡蛋白Bax、抑制肝细胞凋亡而实现。结合相关文献及我们的实验资料,我们认为乙酰化在炎症反应中具有多靶点、多样化的复杂调节作用,其对炎症总体结局的影响还存在许多尚未揭示的新规律,有待进一步的深入细致研究。此外,HATs抑制剂在爆发性肝炎中通过抑制凋亡而发挥保肝效应,这对重症肝炎防治有一定启示意义,但这也需要开展进一步的机制探讨和疗效评估。
[Abstract]:Objective : Acetylation modification is one of the most important methods of post - translationalmodification . It is an important mechanism to regulate the expression of inflammation - related genes .
Methods : LPS ( 10渭g / kg ) and D - Gal ( 700mg / kg ) were injected intraperitoneally in male BALB / c mice .
Western blot was used to detect the protein content in the liver tissues , and the contents of p53 , p53 , caspase - 3 , Bax and Bcl - 2 protein in liver tissues were detected by Western blot .
After formalin fixation , routine HE staining was used to observe the histopathological changes . TUNEL method was used to detect apoptosis , and the level of caspase - 3 was detected by immunohistochemistry .
Results : The results showed that the effect of gartner on hepatic injury induced by LPS / D - Gal was significantly reduced , but the apoptosis of hepatocytes was significantly inhibited by gartner . The results showed that : ( 1 ) The dose - dependent inhibition of the increase of ALT and AST in the plasma induced by LPS / D - Gal was inhibited by Gardiner in the range of 2.5 锝
