人参平肺方对二氧化硅所致大鼠肺纤维化的作用及机制探讨

【摘要】:目的在二氧化硅粉尘悬液致大鼠纤维化模型上观察人参平肺方的作用并探讨其可能机制。方法 SD大鼠麻醉后在气管内注入二氧化硅粉尘悬液以建立肺纤维化模型,然后随机分为模型组、泼尼松组(5.0mg/kg)、人参平肺方3.0、6.0、12.0g/kg剂量组,同时另设空白对照组。连续给药48d后取大鼠肺组织测定肺系数,HE及Masson染色观察肺泡炎症及纤维化程度,测试盒检测肺组织中羟脯氨酸(HYP)含量,ELISA法检测肺泡灌洗液TGF-β1、NF-κB、PDGF、TNF-α及IFN-γ水平,免疫组织化学染色检测肺组织TGF-β1、Samd2/3及ERK1/2蛋白的表达。结果人参平肺方能够显著降低肺纤维化大鼠的肺系数(P0.05),降低肺泡炎症及纤维化程度,降低肺泡灌洗液TGF-β1、NF-κB、PDGF及TNF-α水平(P0.05),升高肺泡灌洗液IFN-γ水平(P0.05),降低肺组织TGF-β1、Samd2/3及ERK1/2的蛋白表达(P0.05)。结论人参平肺方对肺纤维化大鼠有治疗作用,其机制可能与TGF-β/Smad/ERK信号通路有关。
[Abstract]:Objective to observe the effect of Ginseng Pingfei recipe on the rat model of fibrosis induced by silica dust suspension and to explore its possible mechanism.Methods the model of pulmonary fibrosis was established by injecting silica dust suspension into the trachea of SD rats after anesthesia. The rats were randomly divided into two groups: model group, prednisone group (5.0 mg / kg), ginsenpingfei group (3.0 ~ 6.0 ~ 12.0 g / kg) and blank control group.After 48 days of continuous administration, lung tissue was taken from rats to measure lung coefficient, HE and Masson staining to observe the degree of alveolar inflammation and fibrosis. The content of hydroxyproline in lung tissue and the levels of TGF- 尾 1, NF- 魏 -PDGF- 伪 and IFN- 纬 in alveolar lavage fluid were detected by Elisa and the test box was used to detect the level of TGF- 尾 _ 1NF- 魏 -PDGF- 伪 and IFN- 纬 in alveolar lavage fluid.Immunohistochemical staining was used to detect the expression of TGF- 尾 1, Samd 2 / 3 and ERK1/2 protein in lung tissue.Results Ginseng Pingfei recipe could significantly decrease the lung coefficient of rats with pulmonary fibrosis (P0.05), reduce the degree of alveolar inflammation and fibrosis, decrease the levels of TGF- 尾 1, NF- 魏 B, PDGF and TNF- 伪 in alveolar lavage fluid, increase the level of IFN- 纬 in alveolar lavage fluid, and decrease the expression of TGF- 尾 _ 1 Samd _ 2 / 3 and ERK1/2 protein in lung tissue.Conclusion Ginseng Pingfei recipe has therapeutic effect on rats with pulmonary fibrosis, and its mechanism may be related to TGF- 尾 / Smad-ERK signaling pathway.
