低声压级水平次声对未孕大鼠卵巢及胚胎和仔鼠影响的研究

【摘要】:研究目的 本研究通过观察大鼠卵巢的结构及测定雌二醇(E2)、睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)的激素水平以探讨低声压级水平次声波(2~25Hz/70dB)短时间暴露对未孕SD大鼠卵巢功能的影响;通过测定雌三醇(E3)、孕酮(P)的激素水平及记录仔鼠的数量、体质量、身长、尾长、出生时的存活率、3天存活率、性别比等相关指标以探讨低声压级水平次声波短时间暴露对孕鼠的胚胎和仔鼠的影响。 研究方法 将40只SD雌性大鼠和6只SD雄性大鼠饲养两天适应后随机分组:实验1,对照组A1与低声压级水平次声处理组A2各8只,低声压级水平次声波处理方法为30分钟/次×2次/天×20天,于第21天麻醉解剖两组大鼠后取卵巢及血液并处理,卵巢组织经HE染色后在光学显微镜下观察其结构并通过放射免疫分析法测定血清中的雌二醇(E2)、睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)的激素水平;实验2,将24只雌鼠与6只雄鼠按4:1定时合笼,记录每只孕鼠妊娠时间,并将18只孕鼠随机分为对照组B1、妊娠期低声压级水平次声处理组B2(30分钟/次×2次/天×妊娠第1至20天)、妊娠中晚期低声压级水平次声处理组B3(30分钟/次×2次/天×妊娠第7至20天)各6只,,分别于妊娠第6、12、18天经眼眶静脉采血并处理,通过酶联免疫法测血清中的雌三醇(E3)、孕酮(P)的激素水平,待孕鼠分娩后记录仔鼠的数量、身长、尾长、体质量、出生时存活率、3天存活率、性别比等相关指标。 结果 实验1:对照组A1和低声压级水平次声处理组A2大鼠血清中雌二醇(E2)、睾酮(T)、卵泡刺激素(FSH)、黄体生成素(LH)的激素水平的差异无统计学意义(P>0.05);与A1组相比,A2组卵泡的数量及形态无明显改变,但髓质层血管明显扩张。 实验2:对照组B1、妊娠期低声压级水平次声处理组B2和妊娠中晚期低声压级水平次声处理组B3在不同孕期大鼠血清中雌三醇(E3)、孕酮(P)的激素水平的差异均无统计学意义(P>0.05);B1、B2及B3组仔鼠的数量、身长、尾长、出生存活率、3天存活率、性别比无明显差异(P>0.05),与B1组相比, B2、B3组仔鼠的体质量明显增加(P<0.05),B2与B3组仔鼠的体质量无明显差异(P>0.05)。 结论 1.低声压级水平次声短时间作用可以增加大鼠卵巢的血液供应,对卵泡的发育、卵巢的内分泌功能无明显影响; 2.低声压级水平次声短时间作用对孕鼠的激素水平无明显影响,即对胚胎无明显影响,故推测短时间的低声压级水平次声用于孕妇是安全的; 3.低声压级水平次声短时间作用可以增加仔鼠的体质量,对仔鼠数量、身长、尾长、出生时存活率、3天存活率、性别比无明显影响,故推测短时间的低声压级水平次声波暴露对胎儿是安全的。
[Abstract]:Research purpose In this study, we observed the structure of rat ovary and measured the hormone levels of estradiol E _ (2), testosterone, follicle stimulating hormone (FSH) and luteinizing hormone (LH) in order to investigate the effects of infrasound at low sound pressure level on ovarian function in unpregnant SD rats after exposure to infrasonic waves of 25 Hz / 70 dB for a short time. By measuring the hormone levels of estriol E _ 3H, P) and recording the number, body weight, body length, tail length and 3-day survival rate at birth of young rats, Sex ratio and other related indexes were used to study the effects of infrasound exposure at low sound pressure level on embryo and offspring of pregnant rats. Research method Forty SD female rats and six SD male rats were fed for two days and were randomly divided into two groups: control group A1 and low sound pressure level infrasound group A 2, respectively. The treatment method of low sound pressure level infrasonic wave was 30 minutes / time 脳 2 times / day 脳 20 days. The ovary and blood of the two groups were taken and processed after anaesthetized dissection on the 21st day. After HE staining, the structure of ovarian tissue was observed under optical microscope. The levels of estradiol E _ (2), T _ (T), FSH and LH in serum were determined by radioimmunoassay. In experiment 2, 24 female mice and 6 male rats were caged at 4:1 to record the pregnancy time of each pregnant mouse. Eighteen pregnant rats were randomly divided into control group B1, infrasound group with low sound pressure level during pregnancy, group B 2o 30 minutes / times 脳 2 times per day 脳 1 to 20 days of pregnancy, infrasound group with low sound pressure level in the middle and third trimester of pregnancy treated with infrasound group for 30 minutes / time 脳 2 times / d. Each of the 6 rats was on the 7th to 20th day of pregnancy, The blood samples were collected from orbital vein on the 18th day of gestation, and the serum levels of estriol E _ (3) and P _ (P) were measured by enzyme-linked immunosorbent assay (Elisa). The number, length, tail length and body weight of the newborn rats were recorded after delivery. The survival rate at birth was 3 days, sex ratio and other related indicators. Result Experiment 1: there was no significant difference in serum estradiol E _ 2, T _ 2, FSH, LH between the control group A1 and the infrasound group A _ 2 in low sound pressure level (P > 0.05), and the number of follicles in A _ 2 group was higher than that in A _ 2 group (P < 0.05). There was no obvious change in quantity and shape. But the medullary layer vessels dilated obviously. Experiment 2: control group B1, infrasound group B2 during pregnancy and infrasound group B 3 in middle and late pregnancy had no significant difference in the levels of estriol E _ 3 and P) in serum of rats with different pregnancy. The number of litter rats in group B _ 1, B _ 2 and B _ 3, P > 0.05, There was no significant difference in sex ratio (P > 0.05) between group B _ 2 and group B _ 1, but there was no significant difference in body weight between group B _ 2 and group B _ 3 (P < 0.05) and group B _ 3 (P > 0.05). Conclusion 1. Infrasound at low sound pressure level could increase the blood supply of rat ovary, but had no obvious effect on follicle development and endocrine function of ovary. 2. Low sound pressure level infrasound had no obvious effect on hormone level of pregnant rats, that is, it had no obvious effect on embryo, so it is safe to use infrasound of low sound pressure level in pregnant women for a short time. 3. The effect of infrasound at low sound pressure level for short time could increase the body mass of young rats, but had no significant effect on the number, length, tail length, survival rate at birth and 3 days survival rate, and the sex ratio was not significantly affected. Therefore, it is speculated that the infrasound exposure of low sound pressure level for a short time is safe for the fetus.
