隔药饼灸对功能性胃肠病(肝郁脾虚型)大鼠脑肠肽Ghrelin、VIP的影响

【摘要】:目的观察隔药饼灸对功能性胃肠病(肝郁脾虚型)大鼠Ghrelin及VIP的影响,探讨隔药饼灸治疗功能性胃肠病的可能作用机制。 方法60只健康SD大鼠雌雄分笼适应性饲养3天,随机分为5组,每组12只:空白对照组(A组)、模型组(B组)、隔药饼灸组(C组)、多潘立酮组(D组)和逍遥散组(E组),除A组正常饲养外,其余4组均采用复合病因造模法(慢性束缚应激+饮食失节+过度疲劳+夹尾+摇晃)造模,连续21天后,再按被试因素施加方法分组处理:A组正常饲养;B组行捆绑束缚30mim,并灌服生理盐水;C组行隔药饼灸30min,并灌服等量生理盐水;D组行捆绑束缚30min,并灌服多潘立酮;E组行捆绑束缚30min,灌服逍遥散;共14天。实验结束,禁食24h,麻醉处死,腹主动脉采血检测VIP含量,取胃窦和结肠组织检测Ghrelin含量,数据分析。 结果1、一般状态:造模前,各组大鼠皮毛光泽,活泼好动,精神状态良好,粪便球形,干湿适中;造模后,模型大鼠毛发散乱、枯黄无光泽,静卧少动,精神状态疲惫且烦躁,大便多稀溏;经过治疗后,隔药饼灸组、多潘立酮组和逍遥散组大鼠皮毛转光泽,活动增多,精神状态好转,大便由稀变干。2、体重和食量:造模后,大鼠的食量减少,食欲减退,体重增长减缓,有统计学意义(P0.05或P0.01)。3、矿场试验:造模前,大鼠活动度较高,经造模,活动度减少,有统计学意义(P0.05或P0.01)。4、血清VIP含量:模型组与空白组比较,大鼠血清中VIP含量升高(P0.05),隔药饼灸组、多潘立酮组和逍遥散组与模型组比较,VIP的含量均降低(P0.05或P0.01)。5、胃窦和结肠组织中Ghrelin蛋白的表达:模型组与空白组比较,大鼠胃窦和结肠组织中Ghrelin蛋白的表达下降(P0.05或P0.01);隔药饼灸组、多潘立酮组和逍遥散组与模型组比较,Ghrelin蛋白的表达上升(P0.01)。 结论隔药饼灸能降低SD大鼠血清VIP含量,增加组织中Ghrelin蛋白的表达,这可能是隔药饼灸治疗功能性胃肠病的机制之一。
[Abstract]:Objective to observe the effect of herbal cake moxibustion on Ghrelin and VIP in rats with functional gastrointestinal disease (liver stagnation and spleen deficiency) and to explore the possible mechanism of drug separated cake moxibustion in the treatment of functional gastrointestinal disease. Methods 60 healthy Sprague-Dawley rats were fed with male and female cages for 3 days and were randomly divided into 5 groups. There were 12 rats in each group: control group (A group), model group (group B), moxibustion group (group C) and group D (group D) and Xiaoyao powder group (group E), except for group A (normal feeding). The other four groups were made by the method of compound etiology (chronic restraint stress diet, excessive fatigue and tail-shaking) for 21 days. Then according to the factors applied to the subjects, they were divided into two groups: control group A, normal feeding group B, group B were treated by binding and binding 30 min, group C were given normal saline for 30 min, group D was given the same amount of normal saline for 30 min, and group E was given domperidone. Bind and bind for 30 minutes, take Xiaoyao San; A total of 14 days. At the end of the experiment, fasting for 24 hours, anesthetized death, abdominal aorta blood samples to detect the content of VIP, gastric antrum and colon tissue to detect the content of Ghrelin, data analysis. Results 1. General state: before modeling, the rats in each group had luster, liveliness, good mental state, globular feces, moderate dryness and wetness. After making the model, the hair of the model rats was scattered, yellow and luster was dull, the rats were still lying with little movement, and the mental state was tired and irritable. After treatment, the rats in the medicated cake moxibustion group, domperidone group and Xiaoyao powder group changed their fur to luster, their activities increased, their mental state improved, their defecation changed from dilute to dry, and their body weight and food intake decreased after making the model. Loss of appetite and slow growth of body weight, with statistical significance (P0.05 or P0.01P0.01U. 3). Field test: before modeling, the activity of rats was higher, and the activity of rats decreased after modeling. There was significant difference between model group and blank group (P 0.05 or P 0.01). The content of serum VIP in model group was higher than that in blank group, and there was no significant difference between the model group and the blank group. Compared with model group, the content of VIP in serum of rats increased P0.05A, the content of Ghrelin in moxibustion group, domperidone group and Xiaoyao powder group decreased P 0.05 or P0.01A. 5. The expression of Ghrelin protein in gastric antrum and colon: compared with blank group, the expression of Ghrelin protein in gastric antrum and colon: the model group was compared with the blank group. The expression of Ghrelin protein in gastric antrum and colon decreased by P0.05 or P0.01, and the expression of ghrelin protein increased in moxibustion group, domperidone group and Xiaoyao powder group compared with model group. Conclusion moxibustion with separated medicine cake can decrease the content of serum VIP and increase the expression of Ghrelin protein in the tissues of SD rats, which may be one of the mechanisms of treating functional gastrointestinal diseases by moxibustion with separated medicine cake.
