补肾活血颗粒对去势大鼠肾虚证的影响及机制研究

【摘要】：目的:探讨补肾活血颗粒对去势大鼠的肾虚症状的改善作用及作用机制,为临床防治骨质疏松症采用补肾治本的原则提供理论依据。方法:24只雌性大鼠单纯随机分为补肾活血组、模型组、假手术组,每组8只。补肾活血组与模型组麻醉后行双侧卵巢切除术,假手术组麻醉后行假手术处理,饲养3个月确认造模成功后各组分别给予补肾活血颗粒、生理盐水、生理盐水灌胃,90 d后进行3 d大鼠的一般情况观察及生理状态的量化检测,然后麻醉下处死大鼠,用电子天平称取垂体、肾、肾上腺、子宫的湿重,采用Elisa法进行ACTH、E2、T3、T4的血清学检测。结果:补肾活血组大鼠的体毛光滑略显稀疏、行动活泼、反应尚可、精神尚佳、食欲可、身体肥胖、略有嗜睡、眼睛明亮、眼角干燥、二便基本正常等表现;活动度、肛温及进食量均高于模型组,存在显著性差异(P0.01),与假手术组无明显差异(P0.05);血压与模型组及假手术组比较无明显差异(P0.05);血清ACTH、E2、T3、T4的水平明显高于模型组,具有显著性差异(P0.01),与假手术组无明显差异(P0.05);垂体、肾上腺、肾的湿重均明显高于模型组,存在显著性差异(P0.01),与假手术组比较无统计学意义(P0.05),但子宫的湿重高于模型组,低于假手术组,均存在显著性差异(P0.05)。结论:补肾活血颗粒能够明显的改善去势大鼠的肾虚症状,并且推测调节下丘脑-垂体-靶腺轴是补肾活血颗粒改善去势大鼠肾虚症状的作用机制。
[Abstract]:Objective: to investigate the effect and mechanism of Bushen Huoxue granule on kidney deficiency in ovariectomized rats, and to provide a theoretical basis for clinical prevention and treatment of osteoporosis. Methods Twenty four female rats were randomly divided into three groups: tonifying kidney and activating blood circulation group, model group and sham operation group, 8 rats in each group. The model group and the model group were given bilateral ovariectomy after anesthesia, and sham operation group was given sham-operation treatment after anesthesia. After 3 months of feeding, each group was given Bushen Huoxue granule and physiological saline respectively. After 90 days of intragastric administration of normal saline, the rats were observed in general condition and measured quantitatively in physiological state for 3 days. Then the rats were killed under anesthesia, and the wet weights of pituitary, kidney, adrenal gland and uterus were taken with electronic balance. The serological detection of ACTHN E _ 2 T _ 3 T _ 4 was carried out by Elisa method. Results: in the Bushen and Huoxue group, the body hair was smooth and sparse, active, reactive, good in spirit, appetite, obesity, slightly drowsiness, bright eyes, dry eyes, normal activity, etc. The anal temperature and food intake were higher than those in the model group (P 0.01), but there was no significant difference between the model group and the sham operation group (P 0.05), the blood pressure was not significantly different from the model group and the sham operation group (P 0.05), and the serum level of ACTHG E _ 2T _ 3 T _ 4 was significantly higher than that of the model group. The wet weight of pituitary, adrenal and kidney were significantly higher than that of model group (P 0.01), but the wet weight of uterus was higher than that of model group (P < 0.01), but the wet weight of uterus was higher than that of model group (P < 0.05). There was significant difference between sham operation group and sham operation group (P 0.05). Conclusion: Bushen Huoxue granule can obviously improve the symptoms of kidney deficiency in ovariectomized rats, and it is speculated that regulating hypothalamus-pituitary-target gland axis is the mechanism of Bushen Huoxue granule to ameliorate the symptoms of kidney deficiency in ovariectomized rats.
