活血化瘀利水方对大鼠急性软组织损伤模型IL-1β、PGE-2影响的实验研究

【摘要】:目的:观察活血化瘀利水方对大鼠急性软组织损伤模型损伤症候指数、组织形态学的影响以及对损伤局部组织中IL-1p、PGE-2含量的影响,探讨活血化瘀利水方对急性软组织损伤的治疗作用和机制,以及该中药复方对急性软组织损伤修复的作用,为其广泛应用于临床提供实验依据。 方法:将60只SD大鼠随机分为空白对照组、模型对照组、阳性对照组(伤科跌打片组)、活血化瘀利水方低剂量组、活血化瘀利水方中剂量组、活血化瘀利水方高剂量组。除空白对照组外,其余各组均运用局部打击法进行造模,造模后第二天开始给药,于造模后第1、3、5、7天分别观察各组大鼠局部损伤症候指数,治疗7天后将所有大鼠麻醉,无菌条件下取打击中心肌肉组织1g,15mg组织进行组织匀浆,取上清液检测其中IL-1β、PGE-2含量。取0.5cm×0.5cm×0.5cm肌肉组织10%甲醛固定后切片HE染色,进行组织形态学观察。 结果:(1)损伤征候指数观察:造模后第1天,各组大鼠损伤症候指数评分无明显差异(P0.05);造模后第3天,与模型组比较,活血化瘀利水方低剂量组损伤症候指数评分无明显差异(P0.05),其余各组均有明显差异(P0.05或P0.01);造模后第5、7天,活血化瘀利水方各剂量组和伤科跌打片组均能明显促进损伤局部肿胀消退,皮下瘀斑和皮下出血的消散,与模型对照组比有明显的差异(P0.05或P0.01)。 (2)组织形态学观察:与模型对照组比较,活血化瘀利水方各剂量组、伤科跌打片组损伤局部组织的炎症明显减轻,肌纤维的修复更迅速,病理学积分明显降低(P0.05或P0.01),活血化瘀利水方各剂量组与伤科跌打片组相比,病理学积分无明显差异(P0.05)。 (3)IL-1β含量检测:各治疗组损伤局部肌肉组织中IL-1β含量与模型组相比均明显降低(P0.05或P0.01);活血化瘀利水方低剂量组IL-1β含量高于伤科跌打片组(P0.05),中、高剂量组含量与伤科跌打片组相比,无明显差异(P0.05)。 (4)PGE-2含量检测:活血化瘀利水方各剂量组均能明显抑制损伤局部肌肉组织中PGE-2含量的升高,与伤科跌打片组相比,无统计学意义(P0.05);活血化瘀利水方各剂量组和伤科跌打片组与模型组和空白组比较均有统计学意义(P0.05或P0.01)。 结论:活血化瘀利水方具有较明显的抗炎消肿,并能较好促进损伤软组织修复的作用。其治疗软组织损伤的疗效机理之一与部分抑制损伤局部组织中炎性细胞因子IL-1β、PGE-2的产生有关。
[Abstract]:Objective: To observe the effect of Huoxue Huayu Li Shui Recipe on the injury syndrome index, histomorphology and the content of IL-1p and PGE-2 in the local tissues of rats with acute soft tissue injury, and to explore the therapeutic effect and mechanism of Huoxue Huayu Li Shui Fang on acute soft tissue injury, as well as the repair of acute soft tissue injury by the Chinese medicine compound. It provides experimental evidence for its wide application in clinic.
Methods: 60 SD rats were randomly divided into the blank control group, the model control group, the positive control group (Department of Traumatology Dieu tablet group), the low dose group of Huoxue Huayu Li Shui Fang, the middle dose group of Huoxue Huayu Li Shui Fang, the high dose group of Huoxue Huayu Li Shui Fang, and the other groups except the blank control group. The local injury syndrome index of the rats in each group was observed at the first 1,3,5,7 day after the model, and all the rats were anesthetized for 7 days after the treatment. The 1g of the muscle tissue in the center was taken under aseptic conditions and the tissue homogenate of the 15mg tissue was carried out. The content of IL-1 beta and PGE-2 was detected by the supernatant. 0.5cm x 0.5cm x 0.5cm muscle tissue was fixed to the HE staining after 10% formaldehyde fixation. Color, histomorphology observation.
Results: (1) observation of injury syndrome index: there was no significant difference in the score of injury syndrome index (P0.05) in each group at first days after modeling (P0.05). There was no significant difference between the model group and the model group at third days after the model group, and there was no significant difference (P0.05) in the low dose group (P0.05 or P0.01) in the other groups. Each dose group of Huoxue Huayu Li Shui Fang and the group of Dieu tablet in Department of Traumatology all could obviously promote the local swelling and decline of the injury, and the subcutaneous ecchymosis and subcutaneous hemorrhage were obviously different from the model control group (P0.05 or P0.01).
(2) observation of histomorphology: compared with the model control group, the inflammation of the local tissue in the Department of Traumatology of Huoxue Huayu Li Shui Fang was obviously reduced, the repair of muscle fiber was more rapid and the integral of pathology was obviously reduced (P0.05 or P0.01). Significant difference (P0.05).
(3) IL-1 beta content detection: the content of IL-1 beta in the injured local muscle tissue of the treatment group was significantly lower than that in the model group (P0.05 or P0.01), and the content of IL-1 beta in the low dose group of Huoxue Huayu Li Shui Fang was higher than that of the Department of Traumatology (P0.05), and there was no significant difference between the high dose group and the Department of Traumatology drop group (P0.05).
(4) PGE-2 content detection: each dose group of Huoxue Huayu Li Shui Fang can obviously inhibit the increase of PGE-2 content in the injured local muscle tissue, and there is no statistical significance compared with the Department of Traumatology Diping group (P0.05), and there are statistical significance (P0.05 or P0.01) in each dose group of Huoxue Huayu Li Shui Fang and the group of the Department of Traumatology to the model group and the blank group.
Conclusion: the prescription of Huoxue Huayu Li Shui has obvious anti inflammatory and detumescence, and can promote the repair of soft tissue. One of the therapeutic mechanisms for the treatment of soft tissue injury is related to the inhibition of the production of inflammatory cytokines IL-1 beta and PGE-2 in the local tissue.
