灵芝孢子粉对亚急性镉中毒大鼠肾脏的保护作用

【摘要】:目的研究灵芝孢子粉(GLS)对亚急性镉中毒大鼠肾脏的保护作用。方法雄性SD大鼠50只,按体重分层后,采用随机数字法分为5组,每组10只,即正常对照组(C组)、单独染镉组(Cd组)和3个给药组(0.1、0.5、1.0 g/kg GLS+CdCl_2,即GCd_L组、GCd_M组、GCd_H组)。除C组外,其它组大鼠每天灌胃CdCl_2(6.5mg/kg),3个给药组每天灌胃灵芝孢子粉悬浊液,C组和Cd组灌等体积生理盐水,持续42d,定期称重并观察动物的一般表现。染毒结束后麻醉、取血、取肾脏;血生化检测肾功能相关指标;原子吸收光谱法(AAS)检测肾组织及其线粒体、微粒体等亚细胞器中镉的含量;肾脏病理HE染色切片观察病理组织学改变。结果与C组比较Cd组肌酐、尿素、尿酸、胱抑素-c均有不同程度升高,给予GLS干预后,各项指标有不同程度降低,GCd_M组、GCd_H组肌酐变化与Cd组比较,差异有统计学意义(P0.05);给予灵芝孢子粉干预后,镉在各亚细胞器中有不同程度降低,差异有统计学意义(P0.05);HE染色显示,单独染镉组肾组织细胞间质充血严重,细胞排列杂乱,细胞边界不清晰,肾小管管腔不明显,核溶解情况严重,而给药组以上情况均有不同程度改善,并且高剂量组效果优于中剂量组。结论灵芝孢子粉对亚急性镉中毒大鼠肾脏有保护作用。
[Abstract]:Objective to study the protective effect of Ganoderma lucidum spore powder (GLS) on the kidney of subacute cadmium poisoning rats. Methods 50 male SD rats were divided into 5 groups according to the weight stratification and 10 rats in each group, that is, the normal control group (group C), the cadmium group (group Cd) and the 3 administration groups (0.1,0.5,1.0 g/kg GLS+CdCl_2, GCd_L, GCd_M, GCd_H group). Outside the group, the rats in the other groups were intragastric CdCl_2 (6.5mg/kg) every day. 3 administration groups were filled with Ganoderma lucidum spore powder suspension daily, C group and Cd group were filled with physiological saline, continuous 42d, regular weighing and observation of the general performance of the animals. After the end of the poisoning, the blood was taken and the kidney was taken; the blood biochemical test of renal function related indexes; atomic absorption spectrometry (AAS) detection. The content of cadmium in renal tissue and its mitochondria, microsome and other subcellular organelles; pathological changes of kidney pathology HE staining section. Results compared with group C, the creatinine, urea, uric acid and cystatin -c were elevated in different degrees. The prognosis of GLS was given to the group of GLS, the indexes were reduced in varying degrees, GCd_M and GCd_H groups were compared with that of the Cd group. The difference was statistically significant (P0.05). The prognosis of Ganoderma lucidum spore powder decreased in varying degrees, and the difference was statistically significant (P0.05). HE staining showed that the renal interstitial cell congestion was serious, the cells were arranged and disorderly, the cell boundary was not clear, the renal tubules were not obvious, and the nuclear dissolution was serious. The effect of the high dose group was better than that of the middle dose group. Conclusion Ganoderma lucidum spore powder has protective effect on the kidney of subacute cadmium poisoning rats.
