鱼腥草联合龙血竭灌胃对UC大鼠血清IL-10、TNF-α及PAF影响的实验研究

【摘要】:目的:通过建立溃疡性结肠炎(Ulcerative colitis,UC)大鼠模型,观察鱼腥草联合龙血竭对UC大鼠血清白细胞介素10(Inerleukin-10,IL-10)、肿瘤坏死因子(Tumor Necrosis Factor-α,TNF-α)、血小板活化因子(Platelet-Activating Factor,PAF)的影响,验证鱼腥草及龙血竭对UC的治疗作用,探讨二者联合应用是否对UC的治疗有协同作用及其作用机理,从而为UC规范化治疗及用药提供依据。方法:将96只雌雄各半的SD大鼠适应性喂养1周后,随机抽取12只为空白组,84只为造模组。造模前,大鼠全部禁食不禁饮24h,自由饮水。予三硝基苯磺酸(TNBS)/乙醇溶液灌肠造模成功后,将造模组的大鼠随机分为7组,即模型组、柳氮磺吡啶(SASP)组、鱼腥草合龙血竭低剂量组、鱼腥草合龙血竭中剂量组、鱼腥草合龙血竭高剂量组、鱼腥草组、龙血竭组。每组分别给予相应的药物灌胃,3ml/次,1次/日,共14天。给药期间观察各组大鼠的一般情况、体重变化及粪便隐血实验,最后一次给药结束后禁食不禁饮24h,腹腔麻醉下心脏采血,检测大鼠血清IL-10、PAF、TNF-α水平,采血结束后颈椎脱位法处死大鼠,取结肠组织观察各组结肠粘膜变化情况,并做病理组织学检查。结果:鱼腥草合龙血竭高剂量组较鱼腥草合龙血竭中剂量组、SASP组、龙血竭组、鱼腥草组、鱼腥草合龙血竭低剂量组、模型组依次一般状态恢复快,脓血便消失早,中剂量组与SASP组疗效相近。联合用药组、SASP组、龙血竭组、鱼腥草组均表现为肠粘膜组织修复明显,充血水肿,糜烂和溃疡基本愈合。UC大鼠血清中IL-10含量较空白组明显降低,TNF-α、PAF明显升高(P0.01);药物灌胃后,鱼腥草联合龙血竭高剂量组中IL-10含量明显升高,TNF-α、PAF明显降低,与模型组、SASP组、鱼腥草联合龙血竭低、中剂量组、鱼腥草组及龙血竭组相比,差异具有显著性(P0.01);鱼腥草联合龙血竭中、低剂量组、鱼腥草组、龙血竭组、SASP组中IL-10含量亦有升高,TNF-α、PAF也有所降低,与模型组相比有显著性差异(P0.01);SASP组血清IL-10、TNF-α、PAF与鱼腥草联合龙血竭中剂量组、鱼腥草组、龙血竭组比较,无显著性差异(P0.05),但与鱼腥草联合龙血竭低剂量组相比,差异具有统计学意义(P0.05);鱼腥草联合龙血竭低剂量组血清IL-10、TNF-α、PAF与中、高剂量组相比,差异具有统计学意义(P0.05),但与鱼腥草组、龙血竭组相比,差异无统计学意义(P0.05)。结论:1.鱼腥草、龙血竭对UC大鼠模型有治疗作用,二药联合用药对UC的治疗具有协同作用,高剂量组尤甚且疗效优于SASP。2.鱼腥草联合龙血竭治疗UC的可能机制:(1)上调抗炎因子IL-10,从而抑制促炎因子,调节肠道细胞平衡,维持正常肠道粘膜免疫调节。(2)下调PAF,改善血小板激活状态,促进组织修复。下调促炎因子TNF-α,减轻结肠组织的急性炎症反应,阻断UC病变中肉芽肿的形成和纤维组织增生,促进溃疡愈合。
[Abstract]:Objective: to observe the effects of Houttuynia cordata combined with Dragon blood on serum interleukin-10 (IL-10), tumor necrosis factor (Tumor Necrosis Factor- 伪 (TNF- 伪), and platelet activating factor (Platelet-Activating) in UC rats by establishing a rat model of ulcerative colitis (UC). To verify the therapeutic effect of Houttuynia cordata and Dragon Blood Draconis on UC, and to explore whether the combined use of Houttuynia cordata and Draconis has synergistic effect on UC and its mechanism, so as to provide the basis for the standardized treatment and medication of UC. Methods: 96 male and female Sprague-Dawley rats were fed adaptively for one week, and 12 SD rats were randomly selected as blank group (n = 84) and model group (n = 84). Before the model was made, all rats fasted and drank freely for 24 hours. After successful enema with (TNBS) / ethanol solution of trinitrobenzenesulfonic acid, the rats in the model group were randomly divided into 7 groups: model group, (SASP) group, low dose group of houttuynia houttuyniae, middle dose group of houttuynia houttuyniae, and middle dose group of houttuynia houttuyniae. Houttuynia cordata Houttuyniae Houttuynia houttuyniae high dose group, houttuynia group, dragon blood group. Each group was given 3 ml / d intragastric administration of corresponding drugs for 14 days. During the course of administration, the changes of body weight and fecal occult blood test were observed in each group. After the last administration, the rats could not stop drinking for 24 hours. The blood was taken from the heart under abdominal anesthesia, and the level of serum IL-10 and PAFNF-TNF- 伪 was detected. The colonic tissue was taken to observe the changes of colonic mucosa and histopathological examination. Results: the high dose group of Houttuynia cordata Houttuynia houttuyniae was faster than that of the middle dose group of Houttuynia houttuyniae in SASP group, the Dragon blood group and the low dose group of Houttuynia houttuyniae. The general state of the model group recovered faster, and the abscess disappeared earlier than that in the model group. The effect of middle dose group was similar to that of SASP group. The results showed that the intestinal mucosal tissue repair was obvious, hyperemia and edema, and the content of IL-10 in the serum of UC rats were significantly lower than those of the blank group (P0.01), and the contents of IL-10 in the serum of the rats were significantly lower than those of the blank group (P0.01), and the contents of IL-10 in the serum of rats with hyperemia and ulcer healing were significantly lower than those of the control group (P0.01). The content of IL-10 in Houttuynia cordata combined with high dose group was significantly higher than that in model group, Houttuynia houttuyniae combined with dragon blood group, middle dose group, Houttuynia cordata group and dragon blood group, compared with model group, Houttuynia cordata combined with dragon blood group. The difference was significant (P0.01), the content of IL-10 in the low dose group, the houttuynia group, and the dragon blood group also increased, and the content of TNF- 伪 was also decreased. There was no significant difference in serum IL-10 TNF- 伪 PAF between SASP group and Houttuynia cordata group, middle dose group, houttuynia group and dragon blood group (P0.05), but there was no significant difference compared with the low dose group of Houttuynia cordata combined with dragon blood exhaustion (P 0.05). The difference was statistically significant (P0.05); the serum IL-10 TNF- 伪 PAF in low dose group was significantly higher than that in high dose group (P0.05), but there was no significant difference compared with Houttuynia group and Dragon blood group (P0.05). Conclusion 1. Houttuynia cordata and Dragon Blood Draconis have therapeutic effect on UC rat model. The combination of two drugs has synergistic effect on UC, especially in the high dose group and the curative effect is better than that of SASP.2. The possible mechanisms of Houttuynia cordata combined with Dragon Blood in the treatment of UC were as follows: (1) upregulation of anti-inflammatory factor IL-10, thus inhibition of pro-inflammatory factor, regulation of intestinal cell balance, maintenance of normal intestinal mucosal immune regulation, (2) down-regulation of PAF, improvement of platelet activation, and promotion of tissue repair. The inflammatory factor TNF- 伪 was down-regulated, the acute inflammatory reaction of colonic tissue was alleviated, granuloma formation and fibrous tissue proliferation in UC were blocked, and ulcer healing was promoted.
