氯胺酮对人肺癌A549细胞生长的影响及相关机制研究
[Abstract]:Aim: to investigate the effect of ketamine and morphine on the growth of human lung cancer cell line A549. Methods: lung cancer cell line A549 was cultured in vitro for 24 hours. After 24 h culture, the cells were randomly divided into three groups: control group (NC group, treated with 0.9%Na Cl), ketamine group (K1K2K2K3 group, treated with ketamine 100200400 渭 g / mL) and morphine group (M1m2M3 group, treated with morphine 2550100 渭 g / mL, respectively). The migration rate of tumor cells was measured in transwell chamber culture system after 24 hours of continuous treatment. Double luciferase reporter gene assay for NF- 魏 B transcriptional activity 48 h later, the inhibitory rate of cell proliferation was determined by MTT assay. Protein expression of NF- 魏 B p65 was detected by Western blot. The expression of IL-1 尾 and m RNA in cells was detected by QPCR. Results compared with NC group, the effect of M1M 2 group was to promote the migration of A549 cells (P0.05) the effect of M3 group was to inhibit the migration of A549 cells (P0.05). Compared with the NC group, the M1M 2 group was able to promote the proliferation of A549 cells (P0.05), the effect of M3 group was not obvious, and the K2K3 group showed no obvious inhibitory effect on the proliferation of A549 cells. Proliferation of A549 cells (P0.05) the expression of m RNA of IL-1 尾 -COX-2 in M1M2M3 group was significantly higher than that in NC group (P0.05). (P0.05) the expression of m RNA of IL-1 尾 -COX-2 was inhibited in K2K3 group (P0.05). 4 the expression of p65 transcription and protein in M1M2M3 group was not obvious compared with that in NC group (P0.05). (P 0.05) the expression of p65 transcription and protein was inhibited in K1 K2K3 group (P0.05). Conclusion: ketamine inhibits the migration and proliferation of human lung adenocarcinoma cell line A549 through NF- 魏 B signaling pathway and its downstream factors IL-1 尾 and COX-2, which may be more suitable for postoperative analgesia in patients with lung cancer.
