Ad-HGF蛛网膜下腔注射后动脉粥样硬化老龄大鼠大脑皮层血流量变化

【摘要】:目的观察携带人肝细胞生长因子基因的重组腺病毒(Ad-HGF)蛛网膜下腔注射后脑动脉粥样硬化老龄大鼠大脑皮层血流量变化。方法 60只大鼠中随机取45只造模,采用高血压合并高脂饲料喂养制备动脉粥样硬化模型。取建模成功的40只大鼠,随机分为模型组、高剂量组、中剂量组、低剂量组。高、中、低剂量组分别给予5×10~8、2.5×10~8、1.25×10~8pfu/10μL的Ad-HGF,药物经小脑延髓池注入至蛛网膜下腔。模型组给予等体积的生理盐水。分别于给药后第10、20、30天抽取各组大鼠脑脊液,采用ELISA法检测脑脊液中的HGF;将各组大鼠以10%水合氯醛腹腔注射麻醉,于大脑中动脉主要供血区采用激光多普勒血流仪测算脑血流量,以血流仪显示的电压皮伏特值(PU)代表大脑皮层血流量。结果低、中、高剂量组Ad-HGF给药后第10、20、30天脑脊液中HGF水平逐渐降低,且均高于模型组(P均0.05)。中、高剂量组给药后相同时间脑脊液中HGF水平高于低剂量组(P均0.05)。低、中、高剂量组Ad-HGF给药后不同时间大鼠大脑皮层血流量均高于模型组(P均0.05)。低、中、高剂量组Ad-HGF给药后第20、30天大鼠大脑皮层血流量高于给药第10天(P均0.05)。中、高剂量组给药后相同时间大脑皮层血流量高于低剂量组(P均0.05)。结论 Ad-HGF蛛网膜下腔注射可提高脑动脉粥样硬化老龄大鼠大脑皮层血流量,作用与给药剂量、给药时间有关。
[Abstract]:Objective to observe the changes of cerebral cortical blood flow after subarachnoid injection of recombinant adenovirus (Ad-HGF) carrying human hepatocyte growth factor gene in aged rats with cerebral atherosclerosis. Methods Forty-five rats were randomly selected and fed with high fat diet to establish atherosclerosis model. Forty rats were randomly divided into three groups: model group, high dose group, middle dose group and low dose group. The high, medium and low dose groups were given 5 脳 10 ~ (8) (2.5 脳 10 ~ (-8) ~ (1.25 脳 10~8pfu/10) 渭 L Ad-HGF, into the subarachnoid space via the cerebellar medulla oblongata cistern. The model group was given saline of equal volume. Cerebrospinal fluid (CSF) was extracted from each group on day 10, 2030 after administration. HGF; in CSF was detected by ELISA method. The rats in each group were anesthetized with 10% chloral hydrate intraperitoneally, and the cerebral blood flow was measured by laser Doppler flow meter in the main blood supply area of the middle cerebral artery. Cortical blood flow was represented by the voltage Pivot value (PU) displayed by the blood flow meter. Results the levels of HGF in cerebrospinal fluid (CSF) in low, medium and high dose groups were gradually decreased at day 10, 20 and 30 after administration of Ad-HGF, and were higher than those in model group (P 0.05). The level of HGF in cerebrospinal fluid of middle and high dose group was higher than that of low dose group at the same time after administration (all P 0. 05). The cortical blood flow in the low, middle and high dose groups was higher than that in the model group at different time after Ad-HGF administration (all P 0. 05). The cerebral cortical blood flow in the low, middle and high dose groups was higher than that in the 10th day after administration of Ad-HGF (P < 0. 05). The cortical blood flow in the middle and high dose group was higher than that in the low dose group at the same time (P 0.05). Conclusion Ad-HGF subarachnoid injection can increase cerebral cortical blood flow in aged rats with atherosclerosis, and the effect is related to dosage and time of administration.
