一贯煎加味防治大鼠酒精性肝炎肝肾阴虚证的动物实验研究

【摘要】:目的在了解相关动物造模实验资料的前提下,通过白酒灌胃,并辅助长期激怒的方法使大鼠酒精性肝炎肝肾阴虚证的动物模型复制成功,检测给不同剂量的一贯煎加味后各组大鼠的血清生化指标,并观察各组大鼠肝脏病理切片的变化,探讨一贯煎加味对大鼠酒精性肝炎肝肾阴虚证的防治作用。 方法1.一贯煎加味预防大鼠酒精性肝炎肝肾阴虚证:40只200±10g雄性SD大鼠,饲养14天使其充分适应环境,随机分为5组,每组8只。包括对照组,模型组,一贯煎加味预防低剂量组,一贯煎加味预防中剂量组,一贯煎加味高剂量组。除对照组外,其余各组大鼠均双后肢束缚,引起大鼠激怒,待激怒1小时后,,对照组按其习性给予相应饲料,同时用符合比例的蒸馏水灌胃;其他组别给予饲料的同时,每天根据大鼠重量相应给予9g/kg剂量的40。白酒进行灌胃,直到42天,除对照组和模型组外,其它各组大鼠在酒精灌胃1h后,使用符合比例的浓缩中药灌入大鼠体内。模型组在酒精灌胃1h后使用符合比例的蒸馏水进行灌胃,坚持灌胃42天。最后一次灌胃后,将饲料和水从鼠笼撤掉约半天的时间,按比例将水合氯醛配置为6%,按照体重计算药量,麻醉大鼠,从腹主动脉取血,并把血分成两部分,一部分血用于测定雌二醇(E2)、睾酮(T)以及肝功各项指标;另一部分血用于测定环磷酸腺苷(cAMP),并取肝脏在生理盐水中反复冲洗,直至没有淤血,将每组大鼠各自的肝脏取其中的一部分组织泡入福尔马林溶液中固定做病理切片。 2.一贯煎加味治疗大鼠酒精性肝炎肝肾阴虚证:将24只250±10g造模成功的雄性SD大鼠随机分为3组,每组8只。包括一贯煎加味治疗低剂量组、一贯煎加味治疗中剂量组、一贯煎加味治疗高剂量组。对照组和模型组按其习性给予相应饲料和水,同时用符合比例的蒸馏水灌胃;其它组在给予饲料的同时,使用符合比例的浓缩中药灌入大鼠体内。坚持灌胃42天。最后一次灌胃后,将饲料和水从鼠笼撤掉约半天的时间,按比例将水合氯醛配置为6%,腹主动脉取血,将血分成两部分,一部分血用于测定雌二醇(E2)、睾酮(T)以及肝功各项指标;另一部分血用于测定环磷酸腺苷(cAMP)。并取肝脏在生理盐水中反复冲洗,直至没有淤血,将每组大鼠各自的肝脏取其中的一部分组织泡入福尔马林溶液中固定做病理切片。 结果一贯煎加味能够对由于40。白酒灌胃配合激怒法引起的大鼠饮食下降、精神欠佳等方面有一定的改善作用;对40。白酒灌胃引起的血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、AST/ALT、总胆红素(TBIL)、γ-谷氨酰转移酶(γ-GT)、碱性磷酸酶(AKP)的升高有一定的防治作用;对激怒法引起的雌二醇(E2)、E2/T、环磷酸腺苷(cAMP)的升高有一定的防治作用;对40。白酒灌胃引起的总蛋白(TP)、白蛋白(Alb)的降低有防治作用;对激怒法引起的睾酮(T)的降低有防治作用;对大鼠因酒精灌胃导致肝细胞损伤有一定的防治作用。并且,一贯煎加味预防大鼠酒精性肝炎肝肾阴虚方面,中剂量和高剂量的预防效果较好;一贯煎加味治疗大鼠酒精性肝炎肝肾阴虚方面,高剂量的治疗效果较好。 结论大鼠酒精性肝炎肝肾阴虚证造模成功;一贯煎加味对酒精性肝炎肝肾阴虚证有一定的预防作用;一贯煎加味对大鼠酒精性肝炎肝肾阴虚证也有不错的治疗效果。
[Abstract]:Objective To make the animal model of alcoholic hepatitis with Yin deficiency of liver and kidney succeed by intragastric administration of liquor and long-term irritation, and to detect the serum biochemical indexes of rats in each group after different doses of consistently decocting and flavoring, and to observe the changes of liver pathological sections of rats in each group. Objective: To investigate the preventive and therapeutic effects of Liwei Decoction on alcoholic liver hepatitis and yin deficiency syndrome in rats.
Methods 1. Gongguan Jiawei Decoction for Preventing Liver and Kidney Yin Deficiency Syndrome in Rats with Alcoholic Hepatitis: 40 male SD rats were fed for 14 days. They were randomly divided into 5 groups, 8 rats in each group. The rats in the other groups were restrained by hind limbs, which caused the rats to be angry. After one hour of anger, the control group was given the corresponding diet according to their habits and at the same time, the rats in the other groups were given the corresponding dosage of 40. The rats in the model group were fed with distilled water for 42 days after one hour of alcohol administration. After the last gastric administration, the feed and water were removed from the cage for about half a day. The chloral hydrate was proportionally allocated to 6%, and the rats in the model group were fed with distilled water for 42 days. The rats were anesthetized by calculating the dosage according to their body weight. The blood was taken from the abdominal aorta and divided into two parts. One part of the blood was used to measure estradiol (E2), testosterone (T) and liver function. The other part of the blood was used to measure cyclic adenosine monophosphate (cAMP) and the liver was washed repeatedly in normal saline until no congestion occurred. Some of the tissues were injected into formalin solution for pathological sections.
2. Treating alcoholic hepatitis of rats with liver and kidney yin deficiency syndrome by "Gongguan Jiawei": 24 male SD rats were randomly divided into 3 groups, 8 rats in each group, including low dose group treated by "Gongguan Jiawei", "Gongguan Jiawei", "Gongguan Jiawei", "Gongguan Jiawei" group treated by "Gongguan Jiawei", "Gongguan Jiawei" group treated by "Gongguan Jiawei". The control group and model group were The rats in the other groups were fed with the same proportion of concentrated Chinese herbs for 42 days. After the last feeding, the feed and water were removed from the cage for about half a day. Chloral hydrate was proportionally allocated to 6%. Blood was taken from the abdominal aorta and divided into two parts. One part of the blood was used to measure estradiol (E2), testosterone (T) and liver function. The other part of the blood was used to measure cyclic adenosine monophosphate (cAMP).
Results Yiguan Jiawei Decoction could improve the diet and mental health of rats caused by 40.Liquor-infusion combined with irritation; 40.Liquor-infused could increase serum alanine aminotransferase (ALT), glutamic oxaloacetic aminotransferase (AST), AST/ALT, total bilirubin (TBIL), gamma-glutamyltransferase (gamma-GT) and alkaline phosphatase (AKP). It has some preventive and therapeutic effects on the elevation of estradiol (E2), E2/T and cAMP induced by irritation, the decrease of total protein (TP) and albumin (Alb) induced by liquor gavage, the decrease of testosterone (T) induced by irritation, and the liver induced by alcohol gavage in rats. In addition, the medium dose and high dose of Gongguan decoction have better preventive effect on liver and kidney yin deficiency of alcoholic hepatitis in rats, and the high dose of Gongguan decoction has better therapeutic effect on liver and kidney yin deficiency of alcoholic hepatitis in rats.
Conclusion The model of liver-kidney Yin deficiency syndrome in rats with alcoholic hepatitis is successful, the modified decoction has a certain preventive effect on liver-kidney Yin deficiency syndrome in alcoholic hepatitis, and the modified decoction has a good therapeutic effect on liver-kidney Yin deficiency syndrome in rats with alcoholic hepatitis.
