健脾祛痰化瘀方药对脾虚痰浊巴马小型猪回肠线粒体能量代谢相关基因干预作用研究

【摘要】:目的:基于PCR array技术观察脾虚痰浊巴马小型猪回肠线粒体能量代谢相关基因差异性表达及健脾祛痰化瘀方药的干预作用。方法:随机将9只健康的广西巴马小型猪分为正常组、模型组和中药治疗组,每组3只,正常组给予普通饲料饲喂,模型组和中药治疗组采用疲劳过度联合高脂饮食饲喂的方法进行脾虚痰浊动脉粥样硬化模型复制,造模24周后,中药治疗组给予健脾祛痰化瘀方药,48周后,观察巴马小型猪行为学变化,呼吸机麻醉后取材,分离冠状动脉,应用PCR array技术观察巴马小型猪回肠线粒体能量代谢相关基因变化。结果:造模24周后,巴马小型猪出现食少且不欲饮食、神疲乏力、倦怠等明显脾虚证候;PCR array结果显示:与空白对照组比较,模型组变化水平在2倍以上,且经中药干预后表达水平异常被明显纠正的基因共10个,分别为:ATP4B、NDUFS2、NDUFS3、NDUFS4、PPA2、SDHB、SDHC、COX17、NDUFA9、NDUFB1。结论:健脾祛痰化瘀方药对脾虚痰浊巴马小型猪的干预作用可能通过对回肠ATP4B、NDUFS2、NDUFS3等与线粒体能量代谢相关基因实现。
[Abstract]:Objective: to observe the differentially expressed genes related to energy metabolism in ileum mitochondria of spleen deficiency phlegm turbid Bama miniature pig and the intervention effect of invigorating spleen eliminating phlegm and removing blood stasis by PCR array technique. Methods: nine healthy Guangxi Bama miniature pigs were randomly divided into normal group, model group and traditional Chinese medicine treatment group with 3 pigs in each group. The model group and traditional Chinese medicine treatment group were fed with excessive fatigue combined with high fat diet to replicate the model of spleen deficiency and phlegm turbid artery atherosclerosis. After 24 weeks of making the model, the traditional Chinese medicine treatment group was given the prescription of invigorating spleen, removing phlegm and removing blood stasis for 48 weeks. The behavioral changes of Bama miniature pigs were observed, and the coronary arteries were isolated after ventilator anesthesia. The changes of genes related to mitochondrial energy metabolism in ileum of Bama miniature pigs were observed by PCR array technique. Results: after 24 weeks of model making, Bama mini-pigs showed obvious spleen deficiency syndrome, such as eating less and not wanting to eat, fatigue and burnout. The results showed that compared with the blank control group, the change level of the model group was more than 2 times. After intervention with Chinese medicine, there were 10 genes whose abnormal expression level was obviously corrected, namely: 1 ATP-4B, NDUFS2, NDUFS3, NDUFS4, NDUFS4PPA2, SDHBSDHCN, COX17NDUFA9, NDUFB1, and NDUFB1, NDUFA9 and NDUFB1 respectively. Conclusion: the intervention effect of invigorating spleen and removing phlegm and removing blood stasis on spleen deficiency and phlegm turbid miniature pig may be realized by ATP4B,NDUFS2,NDUFS3 and other genes related to mitochondrial energy metabolism.
