邵氏“五针法”对哮喘模型大鼠a-SMA及FN表达影响的实验研究

【摘要】:目的 本课题通过针刺哮喘模型大鼠“肺俞”、“大椎”、“风门”穴,观察邵氏“五针法”对各组哮喘模型大鼠肺组织和血清中平滑肌肌动蛋白-a(a-SMA)及纤维连接蛋白(FN)表达的影响以及对肺组织病理变化的影响,探讨邵氏“五针法”治疗哮喘的机理及其对哮喘模型大鼠气道重塑的作用机制,为针灸治疗哮喘提供科学的实验依据。 方法 选取健康雄性清洁级SD大鼠40只(体重140±20g),随机分成空白对照组、模型对照组、药物治疗组和针刺治疗组,每组10只。采用腹腔注射卵蛋白(OVA)致敏,雾化吸入1%的OVA溶液(每日1次,共7次)激发建立哮喘大鼠模型。针刺治疗组于每日激发后针刺哮喘大鼠“肺俞”(双)、“大椎”、“风门”(双)穴治疗;药物治疗组于每日激发后腹腔注射0.05%地塞米松磷酸钠溶液治疗;模型对照组激发后不做特殊处理;空白对照组腹腔注射及雾化吸入时,用生理盐水代替卵蛋白进行操作。末次治疗后6h内水合氯醛麻醉大鼠,取眼球血后脱颈椎处死大鼠,取右中下肺组织,石蜡包埋,制备病理切片,行苏木精-伊红(HE)染色及a-SMA、FN肺组织免疫组化染色和血清ELISA检测,观察大鼠肺组织形态学改变以及a-SMA、FN在哮喘模型大鼠肺组织和血清中的表达。 结果 1.各组大鼠行为体征观察 模型对照组大鼠与空白对照组比较,出现喷嚏,呛咳,流涕,喘息,呼吸急促,精神不振,反应迟钝;空白对照组大鼠行动敏捷,未见异常;药物治疗组、针刺治疗组大鼠喷嚏,呛咳,喘息等症状较模型对照组明显减轻,反应灵敏。 2.各组大鼠肺组织形态学观察 模型对照组大鼠气道黏膜上皮受损脱落,黏膜下层和平滑肌层增生、变形;气道壁增厚,管腔变窄,管腔内大量分泌物;肺间质大量炎症细胞浸润,肺泡壁破裂变形,肺泡间隔结构紊乱;血管增生,血管管壁增厚。药物治疗组、针刺治疗组气道结构基本完整,肺间质炎症细胞浸润较少,肺组织结构基本正常。空白对照组大鼠气道结构完整,无明显脱落和变形,肺间质未见炎症细胞浸润,肺组织形态结构正常。 3.各组大鼠肺组织a-SMA的比较 空白对照组大鼠肺组织中少见胞浆呈棕黄色染色的a-SMA阳性细胞;模型对照组大鼠肺组织内阳性表达明显增多,与空白对照组比较,差异具有统计学意义(P<0.01);针刺治疗组及药物治疗组与模型对照组比较,肺组织内阳性表达明显减少,差异具有统计学意义(P<0.01),但仍高于空白对照组;针刺治疗组与药物治疗组比较,差异具有统计学意义(P<0.01)。 4.各组大鼠肺组织FN的比较 空白对照组大鼠肺组织中少见胞浆呈棕黄色染色的FN阳性细胞;模型对照组大鼠肺组织内阳性表达明显增多,与空白对照组比较,,差异具有统计学意义(P<0.01);针刺治疗组及药物治疗组与模型对照组比较,肺组织内阳性表达明显减少,差异具有统计学意义(P<0.01),但仍高于空白对照组;针刺治疗组与药物治疗组比较,差异具有统计学意义(P<0.01)。 5.各组大鼠血清a-SMA的比较 模型对照组与空白对照组相比,大鼠血清中a-SMA浓度显著增高,差异具有统计学意义(P<0.01);针刺治疗组及药物治疗组与模型对照组比较,a-SMA浓度明显降低,差异具有统计学意义(P<0.01),但仍高于空白对照组;针刺治疗组与药物治疗组比较,差异具有统计学意义(P<0.01)。 6.各组大鼠血清FN的比较 模型对照组与空白对照组相比,大鼠血清中FN浓度显著增高,差异具有统计学意义(P<0.01);针刺治疗组及药物治疗组与模型对照组比较,FN浓度明显降低,差异具有统计学意义(P<0.01),但仍高于空白对照组;针刺治疗组与药物治疗组比较,差异具有统计学意义(P<0.01)。 结论 1.邵氏“五针法”能够有效改善哮喘模型大鼠的症状和行为体征。 2.邵氏“五针法”能够有效改善哮喘模型大鼠的肺组织形态学改变。 3.邵氏“五针法”能够降低哮喘模型大鼠肺组织中a-SMA和FN的阳性表达。 4.邵氏“五针法”能够降低哮喘模型大鼠血清中a-SMA和FN的阳性表达,且二者在各组哮喘模型大鼠肺组织和血清中的表达具有一致性,并呈正相关。 邵氏“五针法”能明显降低哮喘模型大鼠肺组织和血清中a-SMA及FN的阳性表达,改善气道重塑,说明降低a-SMA、FN的阳性表达是邵氏“五针法”治疗哮喘的作用途径之一。
[Abstract]:Purpose The objective of this study is to adopt acupuncture and asthma model.
