姜黄素对糖尿病大鼠肺组织损伤的保护作用及其可能机制

【摘要】:目的:建立链脲佐菌素(Streptozotocin)诱导的2型糖尿病(Diabetes Melitus)大鼠模型,观察姜黄素(Curcumin)对2型糖尿病大鼠肺组织损伤的保护作用和P38MAPK蛋白表达的影响,探讨其可能的作用机制,为其临床价值提供理论依据。 方法:雄性wistar大鼠随机分为三组:正常对照组(Control)、糖尿病组(DM)、糖尿病+姜黄素治疗组(DM+CUR)。糖尿病组与药物干预组大鼠腹腔注射STZ建立糖尿病模型。成膜后开始给予姜黄素,于15周后测定大鼠血糖和体重,大鼠麻醉处理后,取右肺中叶低温冷冻速冻,同时取右肺下叶置于甲醛溶液中固定。采用苏木素-伊红(HE)染色观察大鼠糖尿病肺组织形态学改变情况。免疫组织化学法(s-p法)和免疫印迹法检测肺组织中p38MAPK和TGF-β1蛋白的表达水平。 结果:1、糖尿病组大鼠与对照组相比血糖显著升高,体重下降。姜黄素组比糖尿病大鼠血糖降低、体重下降缓慢。 2、光镜下正常组肺组织结构清晰,管壁以及粘膜上皮完整。糖尿病组大鼠肺组织结构紊乱,肺泡上皮细胞变性、肺泡腔缩小,肺泡壁结构不完整,间隔毛细血管基底膜增厚、管腔狭窄;纤维结缔组织增生,少许炎细胞浸润。姜黄素治疗组大鼠肺组织损伤较糖尿病组明显减轻。 3、免疫组化(s-p染色)结果显示糖尿病组棕黄色阳性颗粒比对照组多,姜黄素组次之。免疫荧光染色显示p-p38MAPK表达与s-p染色结果相似。免疫蛋白印迹结果糖尿病组与对照组相比肺组织中的p38MAPK及蛋TGF-β1白表达增强(P0.05)。姜黄素组大鼠肺组织中的p38MAPK及TGF-β1蛋白表达较糖尿病组显著下降(P0.05)。 结论:姜黄素可降低糖尿病大鼠血糖,改善糖尿病大鼠体重,减轻糖尿病大鼠肺组织的病理损伤,,其机制可能部分通过影响p38MAPK和TGF-β1蛋白表达有关。
[Abstract]:Aim: to establish a rat model of type 2 diabetic (Diabetes Melitus) induced by streptozotocin (Streptozotocin), to observe the protective effect of curcumin (Curcumin) on lung tissue injury and the expression of P38MAPK protein in type 2 diabetic rats, and to explore its possible mechanism. To provide the theoretical basis for its clinical value. Methods: male wistar rats were randomly divided into three groups: normal control group, (Control), diabetes group, (DM), diabetic curcumin treatment group, (DM CUR). Diabetic model was established by intraperitoneal injection of STZ in diabetic group and drug intervention group. After membrane formation curcumin was given to rats. After 15 weeks blood glucose and body weight were measured. After anesthesia the middle lobe of the right lung was frozen and the lower lobe of the right lung was fixed in formaldehyde solution. The changes of lung histomorphology in diabetic rats were observed by hematoxylin-eosin (HE) staining. The expression of p38MAPK and TGF- 尾 1 protein was detected by immunohistochemistry and Western blotting. Results: 1. Compared with the control group, the diabetic rats significantly increased their blood glucose and lost their body weight. Curcumin group had lower blood glucose and slower weight loss than diabetic rats. 2. The lung tissue structure was clear and the wall and mucosal epithelium were intact in the normal group under light microscope. In the diabetic group, the pulmonary tissue structure was disorder, alveolar epithelial cell degeneration, alveolar cavity shrinking, alveolar wall structure incomplete, capillary basement membrane thickening, lumen stenosis, fibrous connective tissue hyperplasia, a little inflammatory cell infiltration. The lung tissue injury in curcumin treatment group was significantly less than that in diabetic group. 3. The results of immunohistochemistry (s-p staining) showed that there were more brown positive granules in diabetic group than in control group, followed by curcumin group. Immunofluorescence staining showed that the expression of p-p38MAPK was similar to that of s-p staining. Compared with the control group, the expression of p38MAPK and egg TGF- 尾 1 in the diabetic group was higher than that in the control group (P0.05). The expression of p38MAPK and TGF- 尾 1 protein in curcumin group was significantly lower than that in diabetic group (P0.05). Conclusion: curcumin can decrease the blood sugar, improve the body weight and reduce the pathological damage of lung tissue in diabetic rats. The mechanism may be related to the expression of p38MAPK and TGF- 尾 1 protein.
